Continuous flow rapid solid-phase peptide synthesis based on LabVIEW
为实现多肽固相合成的自动化和高效化,结合 LabVIEW 编程能力与流动合成技术,开发连续流动快速固相多肽自动合成系统,探索连续流动模式下单因素的优化策略.结果表明,相比于传统固相合成方法,该系统所需反应时间缩短近80%,试剂用量减少 60%,兼具合成效率与经济优势;粗肽纯度高(阿基瑞林纯度达 90.20%).研究结论验证了该系统在短时间内合成高纯度多肽的能力,为多肽类药物开发及商业化生产提供可靠技术支撑.
In order to achieve automation and high efficiency in peptide solid-phase synthesis,a continuous-flow rapid solid-phase peptide synthesis system was developed.This system was developed by combining the programming capability of LabVIEW and the flow synthesis technology.In addition,the optimization strategy of single factor in continuous-flow mode was explored.The results demonstrated that,in comparison with the conventional solid-phase synthesis approach,the system shortened the reaction time by nearly 80%and decreased the reagents consumption by 60%,thereby exhibiting high synthetic efficiency and economic advantages.The crude peptide purity was found to be 90.20%for Argireline.The findings of this study demonstrated the capacity of the system to synthesize high-purity peptides in a brief period,thereby providing dependable technical support for the development and commercialization of peptide drugs.
