双酶酶解联合乳酸菌发酵黑水虻粗提多肽体外抗氧化活性比较分析OA
Comparative analysis of in vitro antioxidant activity of crude BSF peptides prepared by dual-enzymatic hydrolysis coupled with Lactic Acid Bacteria Fermentation
旨在比较双酶酶解及双酶酶解后分别联合 3 种不同乳酸菌发酵黑水虻制备多肽的体外抗氧化活性.试验分4 组,分别为双酶酶解未接菌组(对照组)、试验 1 组(酶解复合植物乳杆菌发酵组)、试验 2 组(酶解复合粪肠球菌发酵组)、试验 3 组(酶解复合嗜酸乳杆菌发酵组),每组 3 个重复.酶解和发酵产物经离心、过分子筛超滤和超滤后冷冻干燥制备粗提多肽,以维生素 C(Vc)作为参照,对提取的 4 种多肽进行体外抗氧化活性检测.结果表明,与对照组相比,3 个酶菌复合发酵组粗提多肽的总还原力、羟自由基清除能力、超氧阴离子清除能力和 DPPH 自由基清除能力均有显著提升(P<0.05),其中植物乳杆菌和嗜酸乳杆菌发酵组的粗提多肽浓度达到 25 mg/mL 时,与1 mg/mL Vc 的总还原力相近.在大多数试验浓度条件下,植物乳杆菌和嗜酸乳杆菌发酵组的粗提多肽的总还原力、羟自由基清除能力、超氧阴离子清除能力和 DPPH 自由基清除能力显著高于粪肠球菌组(P<0.05).
To evaluate the in vitro antioxidant activity of polypeptides prepared from black soldier fly(Hermetia illucens)processed by enzymatic hydrolysis alone or by enzymatic hydrolysis combined with fermentation using three lactic acid bacteria,four treatments were established with three replicates each:a dual-enzyme(alkaline protease and flavourzyme)hydrolysis group(control),group 1(hydrolysis combined with Lactobacillus plantarum fermentation),group 2(hydrolysis combined with Enterococcus faecalis fermentation),and group 3(hydrolysis combined with Lactobacillus acidophilus fermentation).After enzymatic hydrolysis and fermentation,the products were subjected to centrifugation,molecular-weight cut-off ultrafiltration,and freeze drying to obtain crude polypeptide extracts.Vitamin C(Vc)was used as a reference,and the antioxidant activities of the four crude polypeptide extracts were assessed in vitro.The results showed that,compared with the control,crude polypeptides from all three enzyme-microbe co-fermentation groups exhibited significantly higher total reducing power,hydroxyl radical scavenging activity,superoxide anion scavenging activity,and DPPH radical scavenging activity(P<0.05).Notably,when the crude polypeptide concentration reached 25 mg/mL,the total reducing power of the extracts from group 1 and group 3 was comparable to that of 1 mg/mL Vc.Moreover,at most tested concentrations,crude polypeptides from group 1 and group 3 showed significantly greater total reducing power and radical scavenging activities than those from group 2(P<0.05).These findings provide a basis for developing black soldier fly-derived functional feed additives.
