白花蛇舌草提取物调控TGF-β/Smad信号通路对肾癌细胞增殖、凋亡的影响OA

目的 探究白花蛇舌草提取物对肾癌细胞增殖、凋亡的影响及其机制.方法 将32只接种肾癌Renca细胞的BALB/c小鼠随机分为4组,分别为模型(Model)组、白花蛇舌草提取物(Hedyotis diffusa Willd)组、5-氟尿嘧啶(5-FU)组、TGF-β/Smad通路抑制剂(LY2157299)组,每组8只.相应剂量灌胃给药,模型组给予等体积生理盐水灌胃,1次/d,用药14 d.记录小鼠移植瘤块体积变化,末次给药24 h后处死各组小鼠,称量肿瘤质量,计算抑瘤率;通过Western blot检测TGF-β1、p-Smad2、p-Smad3、Bcl-xl、Bad、cleaved-Caspase-3表达情况;免疫组化法检测增殖细胞核抗原Ki67、PCNA表达水平;肿瘤组织行HE和TUNEL染色检测肿瘤细胞形态和凋亡情况.结果 与模型组比较,3个实验组的移植瘤生长速度均被抑制,给药14 d后的移植瘤体积均明显减小.Western blot结果显示,与模型组比较,3个实验组上调促凋亡蛋白Bad、cleaved-Caspase-3的表达量,下调抗凋亡蛋白Bcl-xl及通路蛋白TGF-β1、p-Smad2、p-Smad3的表达(P<0.05);免疫组织化学结果表明,3个实验组肿瘤组织的Ki67与PCNA的表达显著低于模型组(P<0.05);HE染色和TUNEL染色结果显示:与模型组比较,3个实验组肿瘤组织中的肾癌细胞增殖能力受到抑制,凋亡率显著增加(P<0.05).结论 白花蛇舌草提取物可以抑制肾癌Renca细胞增殖,促进肾癌Renca细胞凋亡,其机制可能与抑制TGF-β/Smad信号通路活化有关.

Objective To investigate the effects and mechanisms of extracts from Baihuasheshecao(Hedyotis diffusa Willd)on the proliferation and apoptosis of renal cancer cells.Method Thirty-two BALB/c mice inoculated with renal cancer Renca cells were randomly divided into model group,Baihuasheshecao(Hedyotis diffusa Willd)extract group,5-fluorouracil(5-FU)group and transforming growth factor-β(TGF-β)/Smad pathway inhibitor(LY2157299)group,with 8 mice in each group.Corre-sponding doses were administered by gavage,and the model group was given an equal volume of physiological saline by gavage once a day for 14 days.The volume changes of transplanted tumor blocks in mice were recorded.The mice in each group were euthanized 24 hours after the last administration.The tumor mass was weighed,and the tumor inhibition rate was calculated.Western blot was used to detect the expressions of TGF-β 1,p-Smad2,p-Smad3,Bcl-xl,Bad and cleaned-Caspase-3.Immunohistochemical method was used to detect the protein expressions of proliferating cell nuclear antigen Ki67 and PCNA.HE and TUNEL staining were performed on tumor tissues to detect the morphology and apoptosis of tumor cells.Result Compared with the model group,the growth rate of transplanted tumors in all three experimental groups was inhibited,and the volume of transplanted tumors significantly decreased after 14 days of administration.Western blot results showed that compared with the model group,the three experimental groups up-regulated the expression levels of pro apoptotic proteins Bad and cleaned-Caspase-3,down-regulated the expressions of anti apoptotic protein Bel-xl and pathway proteins TGF-β1,p-Smad2 and p-Smad3(P<0.05).The immunohistochemical results showed that the expressions of Ki67 and PCNA in tumor tissues of the three experimental groups were significantly lower than those of the model group(P<0.05).The results of HE staining and TUNEL staining showed that compared with those of the model group,the proliferation ability of renal cancer cells in the tumor tissues of the three experimental groups was inhibited,and the apoptosis rate was significantly increased(P<0.05).Conclusion The extract of Baihuasheshecao(Hedyotis diffusa Willd)can inhibit the proliferation of renal cancer Renca cells and promote their apoptosis,which may be related to the inhibition of TGF-β/Smad signaling pathway activation.
