萝卜硫苷对乙型病毒性肝炎小鼠模型的影响及机制研究OA
Effect of Glucoraphanin on Immune-Inflammatory Responses in Mouse Model of Hepatitis B Virus and Its Mechanism
目的:本研究旨在探讨萝卜硫苷对乙型肝炎病毒(HBV)携带小鼠 HBV 复制及免疫调节的影响,并初步探讨其作用机制.方法:采用高压水动力法建立 HBV 转染小鼠模型.将小鼠随机分为正常对照组、模型对照组、萝卜硫苷10、25、50 mg/kg 组.采用 ELISA 法检测血清中乙肝表面抗原(HBsAg)、乙肝 e 抗原(HBeAg)、γ-干扰素(IFN-γ)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活力或含量及乙型肝炎病毒脱氧核糖核酸(HBV DNA)载量;采用免疫组化法检测肝组织中 HBcAg 蛋白表达;流式细胞术检测脾淋巴细胞中 CD4+IFN-γ+T 细胞比例;Western blot 法检测肝组织中 Toll 样受体2(TLR2)、核因子 κB p65(NF-κB p65)、核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)、凋亡相关斑点样蛋白(ASC)、活化半胱天冬酶-1(c-Caspase-1)、白细胞介素-1β(IL-1β)蛋白表达;HE 染色检测肝组织病理变化.结果:相较于正常对照组,模型对照组小鼠血清 HBsAg含量和 HBeAg 活力、HBV DNA 载量、肝组织 HBcAg 蛋白阳性表达明显升高(P<0.05),血清中 IFN-γ 含量、脾淋巴细胞 CD4+IFN-γ+T 细胞百分率明显降低(P<0.05)、肝组织中 TLR2、NF-κB p65、NLRP3、ASC、c-Caspase-1、IL-1β 蛋白表达明显下调(P<0.05);相较于模型对照组,萝卜硫苷25、50 mg/kg 组小鼠血清HBsAg 含量和HBeAg 活力、HBV DNA 载量、肝组织 HBcAg 蛋白表达明显降低(P<0.05),血清中 IFN-γ 含量、脾淋巴细胞 CD4+IFN-γ+T细胞百分率明显升高(P<0.05)、肝组织中 TLR2、NF-κB p65、NLRP3、ASC、c-Caspase-1、IL-1β 蛋白表达明显上调(P<0.05).各组小鼠肝组织细胞结构正常,未观察到明显病理损伤.结论:萝卜硫苷能够发挥抗 HBV 效应,可能与激活 TLR2/NF-κB/NLRP3 通路以增强先天免疫功能有关.
Objective:This study aims to investigate the effect of glucoraphanin on hepatitis B virus(HBV)repli-cation and immunoregulation in HBV-carrying mice and preliminarily explore its mechanism of action.Methods:A mouse model of HBV transfection was established using high-pressure hydrodynamic injection.Mice were randomly di-vided into a normal control group,a model control group,and glucoraphanin treatment groups at doses of 10 mg/kg,25 mg/kg,and 50 mg/kg.Serum levels of hepatitis B surface antigen(HBsAg),hepatitis B e antigen(HBeAg),inter-feron-γ(IFN-γ),alanine aminotransferase(ALT),aspartate aminotransferase(AST),and HBV DNA load were detec-ted by enzyme-linked immunosorbent assay(ELISA).Hepatitis B core antigen(HBcAg)protein expression in liver tis-sue was detected by immunohistochemistry.The proportion of CD4+IFN-γ+T cells in splenic lymphocytes was analyzed by flow cytometry.Western blotting was used to detect the expression of Toll-like receptor 2(TLR2),nuclear factor-κB p65(NF-κB p65),NOD-like receptor protein 3(NLRP3),apoptosis-associated speck-like protein(ASC),cleaved caspase-1(C-caspase-1),and interleukin-1β(IL-1β)in liver tissues.Hematoxylin-eosin(HE)staining was used to de-tect histopathological changes of liver tissue.Results:Compared with the normal control group,mice in the model control group showed significantly increased serum levels of HBsAg,HBeAg,HBV DNA load,and HBcAg protein expression in liver tissues(P<0.05).Meanwhile,serum IFN-γ levels and the proportion of CD4+IFN-γ+T cells in splenic lympho-cytes were decreased(P<0.05),and the expression levels of TLR2,NF-κB p65,NLRP3,ASC,C-caspase-1,and IL-1β proteins in liver tissues were significantly downregulated(P<0.05).Compared with the model control group,mice in the 25 mg/kg and 50 mg/kg glucoraphanin groups showed significantly decreased serum levels of HBsAg and HBeAg,lower HBV DNA load,and reduced expression of HBcAg protein in liver tissues(P<0.05).Additionally,serum IFN-γ levels and the proportion of CD4+IFN-γ+T cells in splenic lymphocytes were increased(P<0.05),and the expression levels of TLR2,NF-κB p65,NLRP3,ASC,C-caspase-1,and IL-1β proteins in liver tissues were significantly upregulated(P<0.05).Histopathological examination revealed normal liver cell structure and no obvious pathological damage in all groups.Conclusion:Glucoraphanin can exert anti-HBV effects,which may be related to the activation of the TLR2/NF-κB/NLRP3 signaling pathway and the enhancement of innate immune function.
