目的:探讨肝郁脾虚型慢性萎缩性胃炎大鼠胃黏膜菌群的结构和丰度变化,同时通过电镜观察胃黏膜的形态改变.通过研究胃黏膜菌群结构的变化,探讨长期服用疏胃方配方药液对大鼠胃内菌群丰度的影响及对胃黏膜的改变.方法:选择经过检疫合格的50 只无特定病原体级SD 雄性大鼠,随机挑选其中10 只作为正常对照组,其余则为模型组.运用以N-甲基-N'-硝基-N-亚硝基胍(N-Methyl-N'-nitro-N-nitrosoguanidine,MNNG)为主的多因素综合方法优化造模,建立肝郁脾虚型萎缩性胃炎的模型.将符合体质量要求的实验大鼠随机分配至模型对照组、胃复春组和疏胃方药液组,每组10 只.正常对照组和模型对照组通过灌胃给予相同体积的纯水,其他各个给药组则以0.9 mL/100 g的剂量每天灌胃给药1次,持续8周.实验完成后,从大鼠的胃窦部获取胃黏膜组织,并制备苏木素-伊红染色法染色的病理切片,然后在电子显微镜下观察每组大鼠的胃黏膜病变情况.提取胃黏膜组织样本,并使用 Illumina HiSeq PE250 高通量测序系统,对样本中的细菌 16S rDNA-V3-V4 区域进行测序.此过程将对胃黏膜微生物群的结构和丰度进行量化分析.结果:疏胃方配方药液组Chao1 丰富度指数、观测物种数指数显著降低;在门水平,疏胃方配方药液组中放线菌门(Actinobacteriota)、拟杆菌门(Bacteroidota)、弯曲杆菌门(Campylobacterota)菌群丰度显著降低(P≤0.05).此外,乳杆菌属(Lactobacillus)菌群丰度显著升高(P≤0.05),单球菌属(Monoglobus)、克里斯滕森氏菌属(Christensenella)菌群丰度显著降低(P≤0.05).结论:疏胃方药液可能通过调节胃黏膜的丰度来改善胃部炎症,从而对慢性萎缩性胃炎起到治疗作用.

Objective:To investigate the structural and abundance changes of the gastric mucosal microbiota in rats with chronic atrophic gastritis(CAG)of liver depression and spleen deficiency type,as well as to observe morphological alterations in the gastric mucosa via electron microscopy.By examining changes in the gastric mucosal microbiota structure,this study explores the effects of long-term administration of the Shuweifang formula liquid(疏胃方配方药液)on the abundance of gastric microbiota and its impact on gastric mucosal morphology in rats.Methods:Fifty specific pathogen-free(SPF)male SD rats,after passing quarantine,were selected,with 10 randomly assigned as the normal control group and the remaining rats designated as the model group.A multifactorial modeling method primarily using MNNG was optimized to establish a rat model of CAG with liver depression and spleen deficiency.The experimental rats meeting weight criteria were then randomly divided into three groups:the model control group,the Weifuchun(胃复春)group,and the Shuweifang formula liguid group,with 10 rats in each group.The normal control and model control groups were administered an equal volume of purified water via gavage,while the other treatment groups received the Shuweifang formula liquid at a dose of 0.9 mL/100 g once daily for 8 weeks.After the experiment,gastric mucosal tissues were collected from the antrum of the rats,and pathological sections were prepared with HE staining.The gastric mucosal lesions in each group were observed under an electron microscope.Gastric mucosal tissue samples were extracted,and the bacterial 16S rDNA-V3-V4 region was sequenced using the Illumina HiSeq PE250 high-throughput sequencing system.This process enabled quantitative analysis of the structure and abundance of the gastric mucosal microbiota.Results:The Shuweifang formula liquid group showed a significant decrease in the Chao1 and Observed species indices.At the phylum level,the abundance of Actinobacteriota,Bacteroidota,and Campylobacterota in the Shuweifang formula liquid group was significantly reduced(P≤0.05).Additionally the abundance of Lactobacillus was significantly increased(P≤0.05),the abundance of Monoglobus and Christensenella was significantly decreased(P≤0.05).Conclusion:The Shuweifang formula liquid may improve gastric inflammation by modulating the abundance of the gastric mucosal microbiota,thereby exerting a therapeutic effect on CAG.
