基于网络药理学与分子对接探究大承气汤治疗脓毒症胃肠功能障碍的作用机制OA
Exploration of the mechanisms of the Da Cheng Qi decoction in treating sepsis-associated gastrointestinal dysfunction based on network pharmacology and molecular docking
目的:通过网络药理学和分子对接方法探讨大承气汤治疗脓毒症胃肠道功能障碍(sepsis-associated gastrointestinal dysfunction,SAGD)的作用机制.方法:通过中药系统药理学数据库与分析平台(TCMSP)筛选大承气汤中四种中药的活性成分及靶标信息.从 GeneCards 数据库中检索 SAGD 相关靶标.通过比较分析确定药物与疾病的交集靶标.使用 STRING 平台构建蛋白质-蛋白质相互作用网络,并将数据导入 Cytoscape 3.7.2 软件.进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析,然后进行关键活性成分与药物靶标之间的分子对接.结果:分析确定了138 个潜在药物靶点及3 379 个疾病靶点,其中 96 个靶点存在重叠.研究表明,大承气汤可能通过包括松弛素信号通路在内的途径发挥其治疗作用.分子对接验证表明,肿瘤蛋白 p53(tumor protein p53,TP53)和芦荟大黄素之间存在良好的结合活性,半胱氨酸天冬氨酸蛋白酶 3(cysteine-dependent aspartate-directed protease 3,CASP3)和川陈皮素之间也存在良好的结合活性.结论:通过对大承气汤和 SAGD 相关靶点的筛选和比较分析,网络分析显示,大承气汤在治疗 SAGD 中表现出多靶点、多基因和多途径的作用机制.除了减轻炎性反应外,它还能维持代谢平衡并预防癌症发展,为SAGD 治疗提供了新方向.
Objective:This study aims to investigate the therapeutic mechanisms of the Da Cheng Qi decoction(大承气汤,DCQD)in treating sepsis-associated gastrointestinal dysfunction(SAGD)using network pharmacology and molecular docking.Methods:The target information of the four traditional Chinese medicines in DCQD was screened via the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The intersection targets between drugs and the disease were identified through comparative analysis.A protein-protein interaction(PPI)network was constructed using the STRING platform,and the data were imported into Cytoscape 3.7.2 software.GO and KEGG enrichment analyses were performed,followed by molecular docking of key active components with drug targets.Results:The analysis identified 138 potential drug targets and 3 379 disease targets,with 96 overlapping targets between them.The study revealed that DCQD may exert its therapeutic effects through pathways including the relaxin signaling pathway.Molecular docking validation demonstrated favorable binding activity between TP53 and aloe-emodin,as well as between(CASP3)and nobiletin.Conclusion:Through screening and comparative analysis of DCQD and SAGD-related targets,network analysis revealed that DCQD exhibits a multi-target,multi-gene,and multi-pathway mechanism in the treatment of SAGD.Besides reducing inflammation,it regulates metabolic homeostasis and prevents cancer development,providing new directions for the treatment of SAGD.
