基于网络药理学与分子对接探索疏脑解郁汤治疗烟雾病伴焦虑抑郁的作用机制OA
Exploring the action mechanisms of the Shu'nao Jieyu decoction in the treatment of moyamoya disease complicated with anxiety and depression based on network pharmacology and molecular docking
目的:运用网络药理学与分子对接技术深入探索疏脑解郁汤治疗烟雾病伴焦虑抑郁的潜在作用机制.方法:借助中药系统药理学数据库与分析平台(TCMSP)筛选疏脑解郁汤的有效活性成分和作用靶点,从 GeneCards 及 OMIM 数据库获取烟雾病伴焦虑抑郁的相关靶点基因,取二者的交集,利用STRING 平台构建蛋白质-蛋白质相互作用网络.再通过CytoScape 搭建"药物成分-靶点"网络并进行拓扑分析,结合 DAVID 数据库完成基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析,对筛选出的核心成分和靶点利用 AutoDock Vina 软件进行分子对接,用 PyMOL 进行可视化展示.结果:疏脑解郁汤作用于烟雾病伴焦虑抑郁的有效成分共有276 个,可作用于 122 个相关靶点基因.KEGG 富集分析主要涉及与蛋白质结合、与腺苷三磷酸结合、蛋白酪氨酸激酶活性等生物信号通路.分子对接显示,刺果甘草查尔酮、川芎哚、异甘草醇、8,6-二甲氧基-5,7,2',5'-四羟基黄酮、莪术醇等核心成分与甘油醛-3-磷酸脱氢酶(glyceraldehyde-3-phosphate dehydrogenase,GAPDH)、肿瘤蛋白 p53(tumor protein p53,TP53)、肿瘤坏死因子(tumor necrosis factor,TNF)、丝氨酸/苏氨酸蛋白激酶 1(Akt serine/threonine kinase 1,AKT1)、白细胞介素(interleukin,IL)-6 等关键靶点结合性良好.结论:本研究初步证实了疏脑解郁汤可通过多成分、多靶点、多途径的整合效应治疗烟雾病伴焦虑抑郁,为后续深入解析其药效物质基础与作用机制提供了理论支撑.
Objective:By using network pharmacology combined with molecular docking,to explore the potential mechanism of the Shu'nao Jieyu decoction(疏脑解郁汤)in treating moyamoya disease complicated with anxiety and depression.Methods:Active ingredients and their corresponding targets of the Shu'nao Jieyu decoction were identified using the TCMSP.Disease-related target genes for moyamoya disease with anxiety and depression were obtained from the GeneCards and OMIM databases.By constructing a Venn diagram,overlapping targets were identified.A protein-protein interaction network was constructed using the STRING platform.A"drug ingredient-target"network was established and subjected to topological analysis using CytoScape software.GO enrichment analysis and KEGG pathway enrichment analysis were performed using the DAVID database.Molecular docking of the screened core ingredients and targets was carried out using AutoDock Vina software,with visualizations presented by PyMOL.Results:the Shu'nao Jieyu decoction was found to act through 276 effective components on 122 related target genes associated with moyamoya disease and anxiety-depression.KEGG enrichment analysis indicated that the regulatory mechanisms primarily involved biological signaling pathways related to protein binding,ATP binding,and protein tyrosine kinase activity.Molecular docking results demonstrated favorable binding affinities between core components(including echinatin,ligustrazine,isoliquiritigenin,8,6-dimethoxy-5,7,2',5'-tetrahydroxyflavone,and curcumol)and key targets such as GAPDH,TP53,TNF,AKT1,and IL-6.Conclusion:Preliminary research has confirmed the Shu'nao Jieyu decoction can treat anxiety and depression associated with moyamoya disease through the integrated effects of multiple components,multiple targets,and multiple pathways,providing theoretical support for further analysis of its pharmacological basis and mechanisms of action.
