温胆汤对脂多糖诱导的RAW264.7巨噬细胞极化及迁移的影响OA
Regulatory effects of the Wendan decoction on lipopolysaccharide-induced polarization and migration in RAW264.7 macrophages
目的:探讨温胆汤含药血清对脂多糖(lipopolysaccharide,LPS)诱导的RAW264.7巨噬细胞极化及迁移的影响.方法:LPS诱导RAW264.7构建巨噬细胞M1型极化模型.细胞计数试剂盒-8(cell counting kit-8,CCK-8)法检测细胞活力,筛选温胆汤含药血清干预巨噬细胞的最佳浓度,然后以温胆汤含药血清干预巨噬细胞,通过巨噬细胞迁移和划痕实验评估细胞迁移能力.酶联免疫吸附试验检测细胞上清液白细胞介素(interleukin,IL)-10,IL-6的水平,实时荧光定量逆转录聚合酶链式反应(real-time fluorescence quantitative reverse transcription polymerase chain reaction,RT-qPCR)法检测IL-6、诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)、IL-10、精氨酸酶-1(arginase-1,Arg-1)的mRNA表达情况.结果:温胆汤含药血清能够减少LPS诱导的RAW264.7巨噬细胞的迁移数量,增加巨噬细胞划痕距离;增加巨噬细胞上清液中IL-10的水平(P<0.05),降低IL-6水平(P<0.05);下调M1型极化标志物IL-6、iNOS的mRNA的表达(P<0.05);上调M2型极化标志物IL-10、Arg-1的mRNA表达(P<0.05).结论:温胆汤能够抑制LPS诱导的巨噬细胞迁移及M1型极化,抑制炎性反应.
Objective:To investigate the effects of serum containing the Wendan decoction(温胆汤)on polarization and migration of RAW264.7 macrophages induced by lipopolysaccharide(LPS).Methods:M1-type polarization model was established by stimulating RAW264.7 macrophages with LPS.Cell viability was assessed using cell counting kit-8(CCK-8)assay to screen the optimal concentration of serum containing Wendan decoction for intervention in macrophages.Subsequently,macrophages were treated with the serum containing Wendan decoction,and their migration ability was evaluated via transwell migration assays and scratch wound assays.The levels of interleukin-10(IL-10)and interleukin-6(IL-6)were measured by ELISA,while mRNA expression of IL-6,iNOS(M1 markers),IL-10,and Arg-1(M2 markers)was quantified using RT-qPCR.Results:Medicated serum containing Wendan decoction can reduces the migration number of RAW264.7 macrophages induced by LPS and increases the scratch closure distance of macrophages(P<0.01).It can increase the levels of IL-10 in the macrophage supernatant(P<0.05)and decrease the levels of IL-6(P<0.05),downregulate the expression of mRNA of IL-6 and iNOS(M1 polarization markers)(P<0.05),upregulate mRNA expression of IL-10 and Arg-1(M2 polarization markers)(P<0.05).Conclusion:Medicated serum containing Wendan decoction could inhibit macrophage migration and M1 polarization induced by LPS,thereby suppressing inflammatory responses.The study suggests its potential as a therapeutic agent for inflammation-related diseases.
