筋骨痛消丸靶向Src通路改善骨关节炎的机制探讨OA
Mechanistic investigation of Jinggu Tongxiao Wan in improving osteoarthritis by targeting the Src pathway
目的:研究筋骨痛消丸改善骨关节炎的作用机制.方法:采用人软骨肉瘤细胞系 SW1353,用脂多糖(lipopolysaccharide,LPS)诱导后分别给予高、中、低浓度筋骨痛消丸含药血清和化学 Src 抑制剂(PP2)处理,通过细胞计数试剂盒-8(cell counting kit-8,CCK-8)、酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)、免疫荧光染色法检测 SW1353 细胞活力、血管内皮生长因子(vascular endothelial growth factor,VEGF)水平和 Src 酪氨酸蛋白激酶(Src protein tyrosine kinase,Src)、Y 染色体性别决定区-盒转录因子[SRY-Box transcription factor 9(sex-determing region of Y chromosome,SRY),SOX9]蛋白表达.结果:LPS 诱导抑制细胞活力,促进VEGF 分泌,上调 Src,而下调 SOX9 表达,筋骨痛消丸含药血清和 PP2 干预则提升细胞活力,抑制 VEGF 分泌,使 Src 表达降低,SOX9 表达相应增加.此外,随着含药血清浓度增加,细胞活力提升,VEGF 水平降低,Src 表达先升高后降低,SOX9 表达上升.结论:筋骨痛消丸含药血清通过下调 VEGF 抑制血管生成,从而抑制Src炎症信号通路并促进SOX9表达,最终抑制骨关节炎.
Objective:To study the mechanism of action of Jinggu Tongxiao Wan(筋骨痛消丸)in ameliorating osteoarthritis.Methods:Human chondrosarcoma cells line SW1353 were induced by lipopolysaccharide(LPS)and then cultured in high,medium and low concentrations of Jinggu Tongxiao Wan drug-containing serum as well as PP2 chemical inhibitors,respectively.Cell viability and its related markers,including VEGF level and expression of SOX9 and Src proteins were detected by CCK-8,ELISA and immunofluorescence staining,respectively.Results:LPS induction inhibited SW1353 cells viability,promoted VEGF secretion,upregulated Src expression,and downregulated SOX9 expression.In contrast,intervention with the drug-containing serum of Jinggu Tongxiao Wan and PP2 enhanced cell viability suppressed VEGF secretion,reduced Src expression,and correspondingly increased SOX9 expression.Moreover,with the increase in the concentration of the drug-containing serum,cell viability was enhanced,VEGF secretion were decreased,Src expression was increased first and then was decreased,and SOX9 expression was elevated.Conclusion:The drug-containing serum of Jinggu Tongxiao Wan inhibits the angiogenesis of blood vessels by suppressing VEGF,thereby blocking the Src signaling pathway and promoting the expression of SOX9,ultimately suppressing osteoarthritis.
