基于网络药理学探讨桂枝茯苓丸治疗子宫肌瘤的作用机制OA
Mechanism of action of Guizhi Fuling Pills in treating uterine fibroids based on network pharmacology
目的 基于网络药理学方法探究桂枝茯苓丸治疗子宫肌瘤的潜在作用机制.方法 利用TCMSP数据库并依据药代动力学参数(OB≥30%,DL≥0.18)筛选桂枝茯苓丸活性成分;通过Pharmmapper、GeneCards数据库分别获取成分和子宫肌瘤的相关疾病靶点.采用Venn图分析得到药物与疾病的交集靶点,借助Cytoscape构建"成分-靶点-通路"关系网络.结果 共筛选出桂枝茯苓丸活性成分76个,作用靶点149个;获得子宫肌瘤疾病靶点937个;药物与疾病的共同作用靶点29个.蛋白质-蛋白质互相作用网络分析得出ALB、ESR1、CASP3、EGFR等核心靶点.基因本体富集分析主要涉及信号转导、对雌二醇的反应等生物过程.京都基因与基因组百科全书通路富集分析提示其作用与雌激素信号通路、MAPK信号通路、癌症通路等密切相关.结论 本研究初步表明桂枝茯苓丸治疗子宫肌瘤体现中药多成分、多靶点、多通路的特点,为其后续深入药理研究提供理论依据.
Objective Investigating the potential mechanism of Guizhi Fuling Pills against uterine fibroids(UF)via network pharmacology.Methods Using the TCMSP database and ADME parameters(OB≥30%,DL≥0.18),the active ingredients of Guizhi Fuling Pills were screened;Predict the target of action of its active ingredients through the Pharmmapper database.Retrieve disease targets related to UF from the GeneCards database.Using Venn diagram to identify the common targets of drug disease interactions,and constructing a"component target pathway"network through Cytoscape.Results As a result,76 active ingredients and 149 targets of Guizhi Fuling Pills were screened out;Obtained 937 targets for UF disease;There are 29 common targets of drug disease interaction.Protein-protein interaction network analysis identified core targets such as ALB,ESR1,CASP3,EGFR,etc.Gene Ontology enrichment analysis mainly involves biological processes such as sighal transduction and response to estradiol.Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis suggests that its effects are closely related to estrogen signaling pathway,MAPK signaling pathway,cancer pathway,and other pathways.Conclusion This study preliminarily indicates that Guizhi Fuling Pills acts on multiple core targets through multiple active ingredients,regulating multiple pathways such as estrogen signaling and MAPK signaling,thereby exerting therapeutic effects on UF.This research providing a theoretical basis for further pharmacological research.
