为进一步综合开发和利用川西小黄菊非药用部位(茎叶),采用HPLC法测定川西小黄菊茎叶中三种黄酮活性成分含量,结果表明三种黄酮类活性成分在川西小黄菊茎叶中均有检出,含量由高到低依次为木犀草素0.2650 mg/g、苜蓿素0.1720 mg/g、芹菜素0.1373 mg/g.同时为评价川西小黄菊茎叶抗炎活性,通过系统溶剂提取法分别制备80%乙醇、石油醚、乙酸乙酯和正丁醇四种提取物.采用CCK-8法检测四种提取物对小鼠单核巨噬细胞的毒性,确定提取物的安全剂量.通过脂多糖诱导小鼠单核巨噬细胞建立炎症模型,采用Griess法测定四种提取物对细胞NO释放量的抑制作用,并用实时荧光定量PCR检测相关炎症因子mRNA表达水平.结果表明川西小黄菊茎叶的正丁醇提取物在50 μg/mL浓度下,无细胞毒性,NO抑制率达到82.59%,推测通过降低TNF-α、IL-1β和iNOS的mRNA表达水平来发挥抗炎作用.研究结果初步表明川西小黄菊非药用部位(茎叶)含有同药用部位(花序)类似的活性成分,存在一定抗炎活性,具备深入开发的价值.

To further develop and utilize the non-medicinal parts(stems and leaves)of Tanacetum tatsienense(T.tatsienense),HPLC method was employed to quantify the contents of three active flavonoids in these parts.The results indicate that all three flavonoids are present in the stems and leaves,with concentrations ranked from highest to lowest as follows:Luteolin at 0.2650 mg/g,tricin at 0.1720 mg/g,and apigenin at 0.1373 mg/g.Additionally,to evaluate the anti-inflammatory activity of the stems and leaves of T.tatsienense,four extracts(80%ethanol,petroleum ether,ethyl acetate,and n-butanol)were prepared using a systematic solvent extraction method.The CCK-8 assay was utilized to assess the toxicity of these extracts on RAW 264.7 cells and to determine their safe dosage.An inflammation model was established by inducing RAW 264.7 cells with LPS.The Griess method was employed to evaluate the inhibitory effects of the extracts on cellular NO release,while q-PCR was used to measure the expression levels of related inflammatory factor mRNAs.The findings revealed that the n-butanol extract of the stems and leaves of T.tatsienense exhibited no cytotoxicity at a concentration of 50 μg/mL,with an NO inhibition rate of 82.59%.This extract demonstrated anti-inflammatory effects by reducing the mRNA expression levels of TNF-α,IL-1β,and iNOS.Overall,the research suggests that the non-medicinal parts(stems and leaves)of T.tatsienense contain active ingredients similar to those found in the medicinal parts(inflorescences),possess notable anti-inflammatory activity,and hold potential for further development.
