基于网络药理学与分子对接探究六妙方治疗脱发的活性成分及靶点验证OA
Investigation of the Active Components and Target Verification of Liumiao Formula Against Alopecia Basesd on Network Phar-macology and Molecular Docking
[目的]采用网络药理学和分子对接技术预测六妙方(Liumiao Formula,LMF)治疗脱发的有效成分及作用靶点,并通过体内实验进行验证.[方法]利用中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)筛选LMF的活性成分及靶点,通过GeneCards、DrugBank和人类孟德尔在线(Online Mendelian Inheritance in Man,OMIM)数据库收集脱发相关基因.取交集后构建蛋白互作(protein-protein interaction,PPI)网络,获得核心成分与靶点;运用DAVID数据库进行基因本体(gene ontology,GO)功能富集分析和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomics,KEGG)通路富集分析;采用Autodock Vina软件进行分子对接验证.构建C57BL/6雄性小鼠脱发模型,采用HE染色和Real-time qPCR验证LMF的生发效果及关键靶点基因表达.[结果]共筛选出LMF的86个活性成分和91个交集靶点;槲皮素、白藜芦醇、山柰酚等为核心成分,与丝氨酸/苏氨酸蛋白激酶(serine/threonine-protein kinase 1,AKT1)、IL-6和TNF-α等核心靶点具有较好的结合活性,结合能均低于-20.9 kJ·mol-1.KEGG分析显示,磷脂酰肌醇3-激酶/丝氨酸/苏氨酸蛋白激酶(phosphatidyl-inositol 3-kinase/serine/threonine protein kinase,PI3K/AKT)信号通路显著富集.体内实验表明,LMF能够显著促进脱发小鼠的毛发再生与毛囊修复,减轻皮肤组织病变.与模型组比较,LMF明显上调皮肤组织AKT1的mRNA表达(P<0.05),降低血清中TNF-α和IL-6的水平(P<0.01,P<0.001).[结论]LMF具有"多成分-多靶点-多通路"协同作用的特点,其活性成分槲皮素、白藜芦醇和山柰酚通过作用于AKT1、IL-6和TNF-α等关键靶点,发挥防脱生发功效.
[Objective]To investigate the active components and therapeutic targets of Liumiao Formula(LMF)against alopecia with network pharmacology and molecular docking,and to verify through in vivo experiments.[Methods]The active components and their targets were retrieved from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP);alopecia-related genes were collected from GeneCards,DrugBank and Online Mendelian Inheritance in Man(OMIM)database.Overlapping targets were used to construct protein-protein interaction(PPI)to screen core compounds and targets.Gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomics(KEGG)pathway enrichment analysis were performed by using the DAVID database.Molecular docking was carried out with AutoDock Vina software.A C57BL/6 mice alopecia model was established,and the hair regenerative effects of LMF as well as the expression of key targets were evaluated by HE staining and Real-time qPCR.[Results]A total of 86 active components and 91 intersection targets were obtained.The active compounds were quercetin,resveratrol and kaempferol,presenting good binding activities with key targets including serine/threonine-protein kinase 1(AKT1),IL-6 and TNF-α,with binding energies less than-20.9 kJ·mol-1.KEGG analysis indicated significant enrichment of the phosphatidylinositol 3-kinase/serine/threonine protein kinase(PI3K/AKT)signaling pathway.In vivo experiments demonstrated that LMF markedly promoted hair regeneration and follicular repair in alopecia model mice,while ameliorating skin tissue lesions.Compared with model group,LMF upregulated the mRNA expression of AKT1 in skin tissue(P<0.05),and decreased serum levels of TNF-α and IL-6(P<0.01,P<0.001).[Conclusion]LMF exhibits characteristics of"multi-component,multi-target and multi-pathway"synergistic effects.Its active components,including quercetin,resveratrol and kaempferol,exert anti-alopecia and hair growth-promoting effects by targeting AKT1,IL-6 and TNF-α.
