OsNramp5基因功能缺失型镉低积累水稻旱育秧赤枯病的发生与防治OA
The Occurrence and Control of Akagare Disease in Dry-raised Seedlings of Low-cadmium Rice with Loss-of-funtion OsNramp5
培育镉低积累水稻品种是解决稻米重金属镉超标经济有效的方法.利用重离子束诱变结合 M1 代靶向深度测序(M1 generation targeted deep sequencing,M1TDS)技术,培育出的 OsNramp5 基因功能缺失型杂交水稻组合低镉臻两优 8612,在长江中下游地区多点试验和示范推广中表现出良好的丰产性和稳定的镉低积累特性.然而,在长江上游的四川德阳示范种植过程中,低镉臻两优 8612 旱育秧出现赤枯症状,具体表现为叶片脉间褪绿并伴有褐色针状斑点,与典型的缺锰(Mn)症状极为相似.为进一步确认其发生赤枯病的原因并提出防治措施,通过苗期水培的方法,比较了对照品种臻两优 8612 和镉低积累水稻品种低镉臻两优 8612、低镉臻两优 3061 对低锰响应的差异.结果表明,在低锰浓度(0.08 μmol/L)条件下,2 个镉低积累水稻品种均表现出与上述赤枯病高度相似的表型,与臻两优 8612 相比,低镉臻两优 8612 的株高、根长、地上部和地下部干重分别显著降低 33.38%、25.38%、56.75%和 65.18%,低镉臻两优 3061 分别显著降低 30.12%、24.25%、56.13%和 63.45%.在水培低锰浓度(0.08 μmol/L)条件下,与不喷施锰叶面肥的处理相比,喷施锰叶面肥 30 000 μmol/L处理的镉低积累水稻品种赤枯病得到明显缓解,其株高、根长、地上部和地下部干重有一定程度的恢复;在旱育秧条件下,播种 14 d 后喷施锰叶面肥 30 000 μmol/L,镉低积累水稻品种赤枯病得到明显缓解,其株高、根长、地上部和地下部干重均恢复到正常水平.综上,低锰条件可诱发镉低积累水稻品种出现赤枯病,喷施锰叶面肥 30 000 μmol/L 处理可明显缓解其赤枯病症状.
Breeding low-cadmium(Cd)rice varieties is an economical and effective approach to address the problem of excessive Cd levels in rice grains.Low-Cd Zhenliangyou 8612,a hybrid rice combination with loss-of-function OsNramp5,has been developed by heavy-ion beam mutagenesis combined with M1 generation targeted deep sequencing(M1TDS)technology.This variety exhibited good yield potential and stable low Cd accumulation characteristic in multi-site trials,demonstrations,and promotions in the middle and lower reaches of the Yangtze River.However,during the demonstration planting process in Deyang of Sichuan Province,located in the upper reaches of the Yangtze River,dry-raised seedlings of low-Cd Zhenliangyou 8612 developed akagare disease symptoms,characterized by interveinal chlorosis and brown needle-like spots on the leaves,which closely resembled typical manganese(Mn)deficiency symptoms.To further confirm the cause of these akagare disease symptoms and propose control measures,this study compared the responses to low Mn conditions between the control Zhenliangyou 8612 and low-Cd rice varieties(low-Cd Zhenliangyou 8612 and low-Cd Zhenliangyou 3061)by hydroponic culture at the seedling stage.The results showed that at a low Mn concentration(0.08 µmol/L),both low-Cd rice varieties exhibited phenotypes highly similar to akagare disease symptoms described above.Compared with Zhenliangyou 8612,low-Cd Zhenliangyou 8612 showed significant decreases of 33.38%,25.38%,56.75%,and 65.18%in the plant height,root length,above-ground dry weight,and below-ground dry weight,respectively.The corresponding significant reductions in low-Cd Zhenliangyou 3061 were 30.12%,24.25%,56.13%,and 63.45%,respectively.Under hydroponic culture conditions with a low Mn concentration(0.08 µmol/L),foliar application of Mn fertilizer at 30 000 μmol/L significantly alleviated akagare disease symptoms in low-Cd rice varieties and partially restored the plant height,root length,and above-ground and below-ground dry weights compared with the treatment without foliar application of Mn fertilizer.For dry-raised seedlings,foliar application of Mn fertilizer at 30 000 μmol/L 14 days after sowing markedly reduced akagare disease symptoms in low-Cd rice varieties,recovering the plant height,root length,and above-ground and below-ground dry weights to normal levels.In conclusion,low Mn conditions can induce akagare disease symptoms in low-cadmium rice varieties,and the treatment with foliar application of Mn fertilizer at 30 000 µmol/L can significantly alleviate akagare disease symptoms.
