水稻花药组织培养特性的细胞质效应研究OA
Cytoplasmic Effects on Tissue Culture Characteristics of Rice Anthers
以一套核置换回交超过 25 代,细胞核均来源于珍汕 97B,细胞质分别来源于饶平野生稻、野败、夜公胞质及珍汕97B 可育胞质的同核异质水稻不育系及保持系材料,与来自日本的粳稻恢复系 Joshu 杂交,构建同核异质材料及其杂交种,通过测定水稻材料在 N6 和 SK3 培养基条件下花药愈伤组织出现时间、诱导率、再生率、绿苗率和白苗率,分析水稻花药组织培养特性的细胞质效应.结果表明,细胞质对 2 种培养基条件下的花药愈伤组织出现时间、诱导率及 N6 培养基条件下的白苗率影响显著或极显著;细胞核与细胞质互作、培养基与细胞质互作效应均对 2 种培养基条件下的花药愈伤组织出现时间、诱导率影响显著或极显著.在 N6 和 SK3 培养基条件下,Joshu 均能缩短花药愈伤组织出现时间,并提高花药愈伤组织诱导率、再生率与绿苗率.在 2 种培养基中,N6 培养基能提高 Joshu 及籼粳杂种 F1 代的花药愈伤组织诱导率、再生率和绿苗率,而 SK3 培养基能缩短花药愈伤组织出现时间.综上,细胞质是影响花药培养效率的重要因素之一,通过筛选特定细胞质与核基因型并优化培养基,可为提高水稻花药培养效率及解析相关遗传机制提供依据.
A set of isonuclear-alloplasmic male sterile and maintainer lines of rice,developed through over 25 generations of nuclear substitution backcrossing,were used in this study.These lines share the same nuclei from Zhenshan 97B but carry different cytoplasms:Raoping wild rice,Yebai,Yegong,and the fertile Zhenshan 97B cytoplasm.They were crossed with the japonica restorer line Joshu from Japan to construct hybrids with identical nuclei but different cytoplasms.The cytoplasmic effects on tissue culture characteristics of rice anthers were analyzed by measuring the callus appearance time,induction rate,regeneration rate,green seedling rate,and white seedling rate of rice anthers under both N6 and SK3 medium conditions.The results demonstrated that cytoplasm had significant or highly significant influences on anther callus appearance time and induction rate under both medium conditions,as well as on the white seedling rate under the N6 medium conditions.Significant or highly significant interaction effects between nucleus and cytoplasm,and between culture medium and cytoplasm on anther callus appearance time and induction rate were observed under both medium conditions.Joshu contributed to shorter appearance time and higher induction,regeneration,and green seedling rates of anther callus under both medium conditions.Between the two media,N6 supported higher anther callus induction,regeneration,and green seedling rates of Joshu and the indica-japonica F1 hybrids,while SK3 was more effective in shortening the anther callus appearance time.In conclusion,cytoplasm is an important factor influencing anther culture efficiency.Selecting appropriate cytoplasmic and nuclear genotypes,along with optimizing the culture medium,can enhance rice anther culture efficiency and provide insights into the underlying genetic mechanisms.
