Research progress on genetic regulation in the development,differentiation and regeneration of vestibular hair cells and gene therapy
前庭毛细胞是内耳前庭系统的核心感受细胞,负责感知头部变速运动并调控身体姿势和协调运动.前庭毛细胞的损伤或丢失是引起多种前庭疾病的主要病理基础.然而,哺乳动物的前庭毛细胞无法自发再生.研究表明,通过基因精细调控和基因治疗可诱导前庭毛细胞再生.基因治疗通过病毒载体递送关键基因至靶细胞,可有效促进毛细胞再生和前庭功能恢复.本文聚焦于影响前庭毛细胞发育、分化及再生过程的关键基因的精细调控机制,并探讨Notch、Wnt、成纤维细胞生长因子等关键信号通路在此过程中所发挥的协同作用,旨在为前庭功能障碍的精准治疗提供参考.
Vestibular hair cells are core vestibular sensory cells in the inner ear,responsible for detecting head acceleration,regulating body posture and facilitating coordinated movement.Damage or loss of vestibular hair cells is a main pathological basis for various vestibular diseases.Vestibular hair cells in mammals are not capable to regenerate spontaneously.Studies have shown that vestibular hair cell regeneration can be induced through targeted genetic regulation and gene therapy.This article focuses on the precise regulatory mechanisms by key genes that affect the development,differentiation,and regeneration of vestibular hair cells,and discusses the synergistic roles of key signaling pathways such as Notch,Wnt,and fibroblast growth factor in this process.Gene therapy,which delivers key genes to target cells via viral vectors,can effectively promote vestibular hair cell regeneration and function recovery.Future research will further explore the optimal strategies,timing,and long-term stability of gene therapy,aiming to pave new ways for developing methods for targeted treatment of vestibular dysfunction.
