归脾汤对慢性束缚应激小鼠焦虑样行为的改善作用及机制OA
Effect of Guipi decoction on anxiety-like behaviors of chronic restraint stress mice and its mechanism
目的 探讨中药复方归脾汤对慢性束缚应激(CRS)模型小鼠焦虑样行为的改善作用及其机制.方法 采用超高效液相色谱-四极杆-静电场轨道离子阱质谱对归脾汤水提物中的主要化学成分进行鉴定.雄性C57BL/6J小鼠随机分为对照组、模型组、模型+丁螺环酮5 mg·kg-1组和模型+归脾汤1.09、2.18、4.36 g·kg-1组,连续ig给药16 d.除对照组外,其余各组小鼠每天给药1h后采用CRS法造模,连续14 d.旷场实验、高架十字迷宫实验(第15天)和明暗箱穿梭实验(第16天)评估小鼠的焦虑样行为;HE染色和高尔基染色检测海马神经元的病理变化;免疫组化法检测沉默信息调节因子1(SIRT1)和脑源性神经营养因子(BDNF)阳性表达;Western印迹法检测突触后致密区95(PSD95)、SIRT1、BDNF、酪氨酸激酶受体B(TrkB)蛋白的表达水平及胞外信号调节激酶(ERK)和cAMP反应元件结合蛋白(CREB)的磷酸化水平.结果 与对照组相比,模型组小鼠中心区域运动距离百分比、中心区域停留时间百分比和进入中心区域次数显著减少(旷场实验);开臂运动距离百分比、开臂停留时间百分比和开臂进入次数显著减少(高架十字迷宫实验);明箱活动时间百分比和明暗箱穿梭次数显著减少(明暗箱穿梭实验);海马神经元数量明显减少,且形态不规则,排列稀疏,核固缩;海马中神经元树突数量、长度和交点个数均显著下降;海马中SIRT1、BDNF阳性表达显著降低,且PSD95、SIRT1、BDNF和TrkB表达及ERK和CREB磷酸化水平均显著降低.与模型组相比,模型+归脾汤2.18和4.36 g·kg-1组小鼠中心区域运动距离百分比、进入中心区域次数、开臂运动距离百分比、开臂停留时间百分比、开臂进入次数、明暗箱活动时间百分比和明暗箱穿梭次数显著增加;海马神经元数量恢复,形态规则,排列有序,模型+归脾汤2.18 g·kg-1组海马神经元无明显核固缩;模型+归脾汤1.09、2.18和4.36 g·kg-1组小鼠树突数量、长度、交点个数均有不同程度增加;模型+归脾汤1.09和2.18 g·kg-1组海马SIRT1、BDNF阳性表达显著升高,模型+归脾汤2.18 g·kg-1组PSD95、SIRT1、BDNF和TrkB表达及ERK和CREB磷酸化水平均显著升高.结论 归脾汤可显著改善CRS模型小鼠的焦虑样行为,其作用机制可能与SIRT1/CREB/BDNF通路改善神经突触可塑性有关.
OBJECTIVE To observe the ameliorative effects of Guipi decoction(GP)on anxiety-like behaviors in a chronic restraint stress(CRS)mouse model and to explore its underlying mecha-nisms.METHODS The major chemical components of GP aqueous extract were identified by-ultra-per-formance liquid chromatography coupled with quadrupole-exactive Orbitrap mass spectrometry(UPLC-Q-Exactive Orbitrap MS).Male C57BL/6J mice were randomly divided into control group,model group,model+buspirone 5mg·kg-1 group,and model+GP 1.09,2.18 and 4.36g·kg-1 groups,with continuous intragastric administration for 16 days.Except for the control group,mice in other groups were subjected to the CRS procedure 1 hour after daily administration for 14 consecutive days.Anxiety-like behaviors were assessed using the open field test,elevated plus maze test(day15),and light-dark box test(day16).Pathological changes in hippocampal neurons were observed by HE staining and Golgistaining.The positive expressions of silent information regulator 1(SIRT1)and brain-derived neurotrophic factor(BDNF)were detected by immunohistochemistry.Western blotting was employed to measurethe expression levels of postsynaptic density protein 95(PSD95),SIRT1,BDNF,and tropomyosin receptor kinase B(TrkB),as well as the phosphorylation levels of extracellular signal-regulated kinase(ERK)and cAMP response element-binding protein(CREB).RESULTS Compared with control group,anxiety-like behaviors,neuronal damage,and synaptic plasticity-related proteins in model group were signifi-cantly reduced.Compared with model group,these behavioral and pathological abnormalities in groups treated with model+GP(2.18 and 4.36 g·kg-1)were significantly ameliorated.Furthermore,the expres-sion of PSD95,SIRT1,BDNF,TrkB,and the phosphorylation levels of ERK and CREB in model+GP2.18 g·kg-1 were significantly up-regulated.CONCLUSION Anxiety-like behaviors in CRS model mice were significantly ameliorated by GP,and its mechanism maybe associated with the improvement of synaptic plasticity via the SIRT1/CREB/BDNF signaling pathway.
