酸枣仁皂苷A调节PINK1/Parkin信号通路介导的线粒体自噬对糖尿病大鼠心肌损伤的影响OA
Effects of jujuboside A on myocardial injury in diabetic rats via regulation of PINK1/Parkin signaling pathway-mediated mitochondrial autophagy
目的 探讨酸枣仁皂苷A(JuA)对糖尿病大鼠心肌损伤的影响及机制.方法 将SD雄性大鼠分为对照组、模型组、二甲双胍组、JuA低剂量组、JuA高剂量组及JuA高剂量+Mdivi-1组.除对照组外均制备糖尿病大鼠模型.检测各组大鼠心功能,观察心肌组织病理学变化及纤维化程度,测定氧化应激、自噬相关指标以及心肌组织中PTEN诱导假定激酶1(PINK1)/Parkin信号通路相关蛋白表达.结果 模型组大鼠左心室舒张末期内径和左心室收缩末期内径、血清肌酸激酶同工酶、乳酸脱氢酶水平、心肌组织胶原容积分数、丙二醛、线粒体超氧化物歧化酶水平、自噬小体数量、PINK1蛋白表达水平高于对照组,而左室射血分数、左室缩短分数、心肌组织SOD活性、p62蛋白水平降低(均P<0.05);二甲双胍及JuA可导致上述指标变化趋势发生逆转;Mdivi-1则明显减弱JuA对DCM大鼠心肌损伤的改善作用.结论 JuA可能通过激活PINK1/Parkin信号通路介导线粒体自噬,抑制心肌细胞氧化应激损伤,改善糖尿病大鼠心肌损伤.
Objective To investigate the effects and mechanisms of jujuboside A(JuA)on myocardial injury in rats with diabetes mel-litus(DM).Methods Male Sprague-Dawley(SD)rats were divided into the following groups:control,model,metformin,low-and high-dose JuA,and high-dose JuA+Mdivi-1 groups.With the exception of the control group,DM model rats were prepared for all groups.For each group,we assessed cardiac function;examined myocardial pathology and the degree of fibrosis;and measured oxidative stress,autophagy-related indicators,and the expression of PTEN-induced putative kinase 1(PINK1)/Parkin pathway-associated proteins in myo-cardial tissue.Results Compared with rats in the control group,the left ventricular end-diastolic diameter(LVEDd)and left ventricular end-systolic diameter(LVESd),serum creatine kinase-MB(CK-MB)and lactate dehydrogenase(LDH)levels,myocardial tissue collagen volume fraction(CVF),malondialdehyde(MDA)content,and mitochondrial reactive oxygen species(ROS)levels,autophagosome count,and PINK1 protein expression were all higher in the model group rats,whereas left ventricular ejection fraction(LVEF),left ventricular fractional shortening(LVFS),myocardial tissue superoxide dismutase(SOD)activity,and p62 protein levels were decreased(all P<0.05).The addition of metformin and JuA reversed the trends observed for the aforementioned indicators,whereas Mdivi-1 significantly attenu-ated the protective effects of JuA on myocardial injury in DM rats(P<0.05).Conclusion JuA may mediate mitochondrial autophagy by activating the PINK1/Parkin signaling pathway,inhibiting oxidative stress-induced damage in cardiomyocytes,and ameliorating myocar-dial injury in DM rats.
