诃子提取物调控COX2/PGE2信号通路对子宫肌瘤细胞生物学行为的影响OA
Effect of Medicine Terminalia Fruit extract on the biological behavior of uterine fibroid cells by regulating the COX2/PGE2 signaling pathway
目的 探讨不同浓度诃子提取物对子宫肌瘤细胞生物学行为的影响及机制.方法 将人子宫肌瘤细胞分为对照组,低、中、高浓度(60、90、120 μg/mL)诃子提取物组和高浓度诃子提取物+LPS组(120 μg/mL诃子提取物和3 μg/mL LPS处理).利用CCK-8法检测各组子宫肌瘤细胞增殖能力;流式细胞仪检测各组子宫肌瘤细胞凋亡率;划痕实验检测各组子宫肌瘤细胞迁移能力;Transwell检测各组子宫肌瘤细胞侵袭能力;Western blotting检测各组子宫肌瘤细胞中COX2和PGE2蛋白表达.结果 与对照组相比,诃子提取物处理组子宫肌瘤细胞增殖率、迁移和侵袭能力均显著降低(P<0.05),细胞凋亡率显著升高(P<0.05),子宫肌瘤细胞中COX2和PGE2蛋白表达水平均显著降低(P<0.05);且作用效果随着诃子提取物浓度增加而增强(P<0.05).COX2激活剂LPS能够显著逆转诃子提取物对子宫肌瘤细胞的作用.结论 诃子提取物可显著降低子宫肌瘤细胞增殖、迁移和侵袭能力,并促进细胞凋亡,这种作用可能是通过抑制COX2/PGE2信号通路来实现的.
Objective To investigate the effects and mechanisms of different concentrations of Medicinal Terminal Fruit extracts on the biological behavior of uterine fibroid cells.Methods Human uterine fibroid cells were assigned into control group,low-,medium-,and high-concentration(60,90,and 120 μg/mL)Medicinal Terminal Fruit extract groups,and high-concentration Medicinal Terminal Fruit extract+LPS group(120 μg/mL Medicinal Terminal Fruit extract and 3 μg/mL LPS).The CCK-8 assay was used to detect the proliferative ability of uterine fibroid cells in each group.Flow cytometry was used to detect the apoptosis rate of uterine fibroid cells in each group.The scratch test was used to detect the migratory ability of the uterine fibroid cells in each group.Transwells were used to detect the invasive ability of the uterine fibroid cells in various groups.Western blotting was performed to detect the expression levels of cyclooxygenase-2(COX2)and prostaglandin E2(PGE2)proteins in uterine fibroid cells from various groups.Results Compared with the control group,the proliferation rate,migration rate,and invasive abilities of uterine fibroid cells in each of the Medicinal Terminal Fruit extract groups were significantly reduced(P<0.05),the apoptosis rate was significantly increased(P<0.05),and the expression of COX2 and PGE2 proteins in uterine fibroid cells were significantly decreased(P<0.05).Medicinal Terminal Fruit extract inhibited the malignant behavior of uterine fibroid cells,and this effect increased with increasing extract concentration(P<0.05).However,COX2 activator LPS reversed the effect of Medicinal Terminal Fruit extract on uterine fibroid cells(P<0.05).Conclusion Medicinal Terminal Fruit extract reduced the proliferation,migration,and invasive abilities of uterine fibroid cells and promoted apoptosis.This effect may be achieved by inhibition of the COX2/PGE2 signaling pathway.
