目的 探讨LINC01082通过调节miR-18a-3p/CADM2轴对食管鳞状细胞癌(ESCC)细胞增殖、迁移和侵袭的影响.方法 应用UALCAN数据库分析LINC01082、miR-18a-3p和CADM2在ESCC组织和癌旁组织中的表达;采用实时定量PCR检测LINC01082、miR-18a-3p和CADM2在ESCC细胞和正常食管上皮细胞中的表达;在ESCC细胞系KYSE150中过表达LINC01082,采用CCK-8、平板克隆、划痕实验和Transwell检测LINC01082对KYSE150细胞增殖、迁移和侵袭能力的影响;采用双萤光素酶报告基因实验检测LINC01082与miR-18a-3p、miR-18a-3p与CADM2的相互作用.结果 LINC01082和CADM2在ESCC组织中的表达水平显著低于癌旁组织,miR-18a-3p在ESCC组织中的表达水平显著高于癌旁组织(P<0.05).与正常食管上皮细胞SHEE细胞相比,KYSE150细胞中LINC01082和CADM2 mRNA表达水平降低(P<0.05),而miR-18a-3p mRNA表达水平显著升高(P<0.05).在KYSE150细胞中过表达LINC01082后,细胞增殖、迁移和侵袭能力显著下降.双萤光素酶活性检测结果表明,LINC01082与miR-18a-3p间、miR-18a-3p与CADM2间存在靶向关系.结论 过表达LINC01082可能通过下调miR-18a-3p,促进CADM2表达,从而抑制ESCC细胞的增殖、迁移和侵袭.

Objective To investigate the effects of LINC01082 on the proliferation,migration,and invasion of esophageal squamous cell carcinoma(ESCC)cells by regulating the miR-18a-3p/CADM2 axis.Methods LINC01082,miR-18a-3p,and CADM2 expressions in ESCC tissues and adjacent normal tissues were analyzed using the UALCAN database.LINC01082,miR-18a-3p,and CADM2 mRNA expressions in ESCC cells and normal esophageal epithelial cells were measured using real-time quantitative PCR.LINC01082 was over-expressed in the ESCC cell line KYSE150,and its effects on the proliferation,migration,and invasion of KYSE150 cells were detected using the cell counting kit(CCK)-8,plate colony formation,scratch,and Transwell assays.A dual luciferase reporter assay was performed to detect the interactions between LINC01082 and miR-18a-3p and between miR-18a-3p and CADM2.Results Compared with those in the adjacent normal tissues,LINC01082 and CADM2 expression levels were significantly lower in ESCC tissues,whereas the miR-18a-3p expression level was significantly higher(P<0.05).Compared with normal esophageal epithelial cells(SHEE cells),LINC01082 and CADM2 mRNA expression levels were significantly decreased in KYSE150 cells,whereas miR-18a-3p expression level was significantly increased(P<0.05).Notably,LINC01082 overexpression significantly reduced cell proliferation,migration,and invasion abilities of KYSE150 cells.The results of dual luciferase reporter assay revealed a targeting interaction between LINC01082 and miR-18a-3p,as well as between miR-18a-3p and CADM2.Conclusion LINC01082 overexpression may inhibit the proliferation,migration,and invasion of ESCC cells by downregulating miR-18a-3p and promoting CADM2 expression.
