嵌合抗原受体T细胞疗法在卵巢癌治疗中的进展、挑战与探索OA
Progress,challenge,and exploration of chimeric antigen receptor T cell therapy for treating ovarian cancer
卵巢癌是最常见的妇科恶性肿瘤之一,具有预后差、死亡率高的特点,被认为是最致命的妇科肿瘤.大多数卵巢癌患者直至晚期才被诊断,导致5年生存率低于50%.近年来,嵌合抗原受体(CAR)T细胞疗法在肿瘤治疗领域迅速发展,成为一种极具前景的卵巢癌治疗手段.然而,其临床转化仍面临诸多挑战,包括严重不良反应、非靶点效应、肿瘤抗原异质性以及免疫抑制性肿瘤微环境等.本文通过总结和分析CAR-T细胞疗法在卵巢癌治疗中的研究现状以及面临的挑战,探讨其潜在的优化策略和未来研究方向.
Ovarian cancer is one of the most prevalent gynecological malignancies.It is characterized by poor prognosis and is one of the gynecological cancers with the highest mortality rates.Most ovarian cancer patients are not diagnosed until the disease reaches an advanced stage,with the 5-year survival rate being<50%.Current treatment for ovarian cancer primarily relies on surgical intervention and chemotherapy.In recent years,immunotherapy has emerged as a novel therapeutic approach for ovarian cancer,among which chi-meric antigen receptor(CAR)-T cell therapy represents a significant advancement in cancer immunotherapy and is a promising immu-notherapeutic strategy.However,this therapy,which has achieved significant success in hematologic malignancies,still faces numerous challenges and obstacles in the context of ovarian cancer treatment.This review summarizes the current research progress and challenges associated with CAR-T cell therapy in ovarian cancer.In addition,strategies to enhance the therapeutic efficacy of CAR-T cell-based interventions are explored.
