组蛋白去乙酰化酶抑制剂对甜菜抽薹开花相关基因表达的影响OA
Effects of Histone Deacetylase Inhibitors on the Expression of Bolting and Flowering-Related Genes in Sugar Beet
[目的]为了系统探究组蛋白去乙酰化酶(HDACs)在甜菜抽薹开花过程中的功能机制,解析 Zn2+依赖型 HDACs 抑制剂对开花基因表达的调控效应.[方法]通过施用 Tubastatin A、Quisinostat 等 HDACs 抑制剂,分析其对甜菜开花相关基因表观修饰状态的影响.[结果]半定量PCR 检测结果显示,Tubastatin A 能明显促进BvFT1 的表达,同时对BvBTC1、BvGI、BvCO 的表达有显著的抑制作用;Quisinostat 对这几个基因的影响不显著;FNDR-20123 显著促进 BvBTC1、BvGI、BvCO 的表达;Resminostat 的施加导致了 BvBTC1 的表达水平显著下调.荧光定量 PCR 结果显示,Tubastatin A 可使 BvBTC1 和 BvGI 的表达水平也显著上调,这与半定量 PCR 检测结果不一致.HDACs 抑制剂可通过抑制酶活性显著改变开花基因的组蛋白乙酰化水平,进而调控甜菜抽薹开花的时间进程,首次建立 RPD3/HDA1 家族与甜菜开花调控的直接关联.[结论]本研究完善了甜菜开花调控的表观遗传理论体系,阐明了 HDACs 活性与开花基因表达的调控网络,为甜菜早熟育种及抗抽薹品种改良提供了技术路径,对长日照作物开花机制研究具有范式参考价值.
[Objective]This study aimed to systematically explore the functional mechanism of histone deacetylases(HDACs)in the bolting and flowering process of sugar beets,and to analyze the regulatory effects of Zn2+-dependent HDACs inhibitors on the expression of flowering-related genes.[Methods]By applying HDACs inhibitors such as Tubastatin A and Quisinostat,the effects on the epigenetic modification status of beet flowering-related genes were analyzed.[Results]Semi-quantitative PCR results showed that Tubastatin A significantly promoted the expression of BvFT1,while significantly inhibiting the expression of BvBTC1,BvGI,and BvCO;Quisinostat had no significant effect on these genes;FNDR-20123 significantly promoted the expression of BvBTC1,BvGI,and BvCO;and Resminostat application led to a significant down-regulation of BvBTC1 expression levels.Quantitative real-time PCR results showed that Tubastatin A also significantly up-regulated the expression levels of BvBTC1 and BvGI,which was inconsistent with the semi-quantitative PCR results.HDACs inhibitors could significantly alter the histone acetylation levels of flowering genes by inhibiting enzymatic activity,thereby regulating the time course of sugar beet bolting and flowering.This study first established a direct association between the RPD3/HDA1 family and beet flowering regulation.[Conclusion]This study improves the epigenetic theoretical system of sugar beet flowering regulation,clarifies the regulatory network between HDACs activity and flowering gene expression,provides a technical path for early-maturing breeding and bolting-resistant variety improvement of sugar beets,and has paradigmatic reference value for the study of flowering mechanisms in long-day crops.
