为了探索猪流行性腹泻病毒(PEDV)母源抗体的建立方法,本试验对返饲+组织灭活苗与商品苗+组织灭活苗的免疫效果进行对比分析,对返饲母猪乳汁中PEDV免疫球蛋白A(IgA)含量进行衰减分析,测定母猪PEDV黏膜免疫的维持时间,并对组织灭活苗对乳汁PEDV IgA含量的影响进行分析.结果显示,返饲+组织灭活苗的免疫方式可以使母猪产生高水平的IgA抗体,且极显著高于商品苗+组织灭活苗的免疫方式(P<0.01);母猪乳汁中PEDV IgA含量随着哺乳时间增加而不断降低,但仍能够为仔猪提供免疫保护;经产母猪初乳中PEDV IgA含量显著高于后备母猪(P<0.05);母猪经过1次反饲并持续进行组织灭活苗跟胎免疫后,其初乳中PEDV IgA抗体高水平表达可持续5~6胎次之久;反饲后跟胎免疫组织灭活苗2个胎次的母猪初乳PEDV IgA含量与2个胎次前无显著差异(P=0.075),反饲后停止跟胎免疫的母猪,2个胎次后初乳PEDV IgA含量较2个胎次前极显著下降(P<0.01).结果表明,返饲+组织灭活苗跟胎免疫可诱导高水平PEDV IgA抗体,同时组织灭活苗的持续跟胎免疫对维持高水平抗体至关重要.本试验结果为建立高水平PEDV母源抗体提供了参考.

To explore effective methods for establishing maternal antibodies against porcine epidemic diarrhea virus(PEDV),this study compared the immunization effects of feedback exposure plus inactivated tissue vaccine with those of commercial vaccine plus inactivated tissue vaccine.The decay of PEDV immunoglobulin A(IgA)in colostrum and milk of feedback-exposed sows was analyzed,the duration of mucosal immunity was assessed,and the effect of inactivated tissue vaccine on PEDV IgA content in milk was examined.The results showed that the immunization strategy of feedback exposure combined with inactivated tissue vaccine induced significantly higher levels of IgA antibodies in sows than the commercial vaccine plus inactivated tissue vaccine strategy(P<0.01).The content of PEDV IgA in milk decreased gradually with lactation time but still provided immune protection for piglets.PEDV IgA content in colostrum of sows were significantly higher than those in gilts(P<0.05).After a single feedback exposure followed by continuous parity-based immunization with inactivated tissue vaccine,high-level expression of PEDV IgA in colostrum was maintained for 5-6 parities.In sows that received inactivated tissue vaccine immunization for two consecutive parities after feedback exposure,PEDV IgA content in colostrum showed no significant change compared with two parities earlier(P=0.075).However,in sows that ceased parity-based vaccination after feedback exposure,PEDV IgA content in colostrum significantly decreased after two parities(P<0.01).These findings indicate that feedback exposure combined with continuous parity-based inactivated tissue vaccination can effectively induce high levels of PEDV IgA antibodies,and ongoing parity-based immunization is critical for maintaining antibody levels.This study provides a valuable reference for establishing high-level maternal immunity against PEDV.
