犬白介素31蛋白表达及其单克隆抗体制备OA
Expression of canine IL-31 protein and preparation of its monoclonal antibodies
IL-31 通过与其受体结合,在多种皮肤炎症过程中发挥关键生物学作用.本研究通过构建犬IL-31蛋白原核表达载体,利用大肠杆菌表达系统成功表达后纯化出高纯度的重组犬IL-31 蛋白.以该重组蛋白免疫BALB/c小鼠,通过 3 次免疫、细胞融合与有限稀释法筛选,成功获得 1 株可稳定分泌抗犬IL-31 单克隆抗体的杂交瘤细胞株 1F2,其抗体亚型为IgG1/λ,纯化后的抗体效价可达 1∶13 107 200.为进一步验证抗体的特异性,构建真核表达质粒并在HEK-293T细胞中表达犬IL-31.Western-blot结果表明,1F2 单克隆抗体能够特异性识别真核系统中表达的犬IL-31 蛋白,表明有良好的反应特异性.本研究成功制备的抗犬IL-31 单克隆抗体 1F2,为后续开发针对犬IL-31 的治疗性抗体及相关检测方法的建立奠定了基础.
Interleukin-31(IL-31)exerts a key biological role in various cutaneous inflammatory processes by binding to its receptor.In this study,the prokaryotic expression vector for canine IL-31 was constructed,and the recombinant canine IL-31 protein with high purity was successfully expressed and purified using an E.coli expression system.After immunizing BALB/c mice with this recombinant protein,followed by three rounds of immunization,cell fusion,and screening via the limited dilution method,one hybridoma cell line 1F2 that could stably secrete canine IL-31 monoclonal antibody was successfully obtained.The antibody was of the IgG1/λ isotype,and the titer of the purified antibody could reach 1∶13 107 200.To further verify the antibody specificity,a eukaryotic expression plasmid was construct-ed and used to express canine IL-31 in HEK-293T cells.Western-blot results showed that the 1F2 mono-clonal antibody could specifically recognize canine IL-31 protein expressed in the eukaryotic system,in-dicating excellent reaction specificity.The successfully prepared anti-canine IL-31 monoclonal antibody 1F2 in this study lays a foundation for the subsequent development of therapeutic antibodies targeting canine IL-31 and the establishment of related detection methods.
