Oncolytic viruses(OVs),as an innovative strategy for cancer immunotherapy,are increasingly demonstrating sig-nificant potential in cancer treatment due to their high efficacy,precise tumor targeting,and relatively low side effects.This article provides a comprehensive review of the evolution of OV treatment strategies,scientific foundations,genetic engineering modifica-tions,clinical application strategies,and the potential for synergistic effects when used in combination with other therapeutic ap-proaches.It also discusses the challenges faced by OVs in the clinical translation process,including safety,efficacy assessment,and selection of viral vectors.In addition,the article introduces several commercialized OV drugs,such as H101,T-VEC and Delytact,which have shown positive therapeutic effects in the treatment of various types of cancer.The aim of this article is to explore innovative applications of OVs in cancer treatment and the challenges they face,providing a comprehensive perspective for researchers in the fields of immunology and oncology,in order to promote the scientific development and clinical application of this therapy.
