活血滋肾方对子宫内膜异位症大鼠Th17/Treg平衡及炎症因子表达的影响OA
Effect of Huoxue Zishen Decoction on Th17/Treg balance and inflammatory factors expression in endometriosis rats
目的:探究活血滋肾方对子宫内膜异位症(EMs)大鼠辅助性T细胞17/调节性T细胞(Th17/Treg)平衡及炎症因子表达的影响.方法:60只大鼠随机分为假手术(Sham)组、EMs组、低剂量活血滋肾方(HXZS-L)组、高剂量活血滋肾方(HXZS-H)组,每组15只.采用自体子宫内膜移植法制备大鼠EMs模型.Sham组仅结扎并切除左侧子宫角,不移植.造模2周后,HXZS-L组、HXZS-H组大鼠分别每日灌服活血滋肾方煎剂(10.71 g/kg、42.84 g/kg);Sham组和EMs组大鼠灌服等量生理盐水,连续灌服3周.治疗3周后,ELISA检测血清炎症因子IL-6、IL-17A、IL-22、IL-10、TGF-β水平;HE染色观察异位内膜组织损伤;流式细胞术检测外周血Th17/Treg;RT-qPCR、Western blot检测异位内膜组织Th17/Treg相关标志物(IL-17A、RORγt、Foxp3、GITR、CTLA-4)mRNA和蛋白水平.结果:与Sham组相比,EMs组大鼠异位内膜病灶隆起、透亮,周围新生大量血管,异位内膜上皮细胞呈多层紊乱排列,外周血Th17比例、Th17/Treg升高,Treg比例降低,异位内膜组织IL-17A、RORγt mRNA和蛋白水平升高,Foxp3、GITR、CTLA-4 mRNA和蛋白水平降低,血清IL-6、IL-17A、IL-22水平升高,IL-10、TGF-β水平降低(P<0.05);与EMs组相比,HXZS-L组、HXZS-H组大鼠异位内膜病灶缩小,周围新生少量血管,异位内膜上皮细胞呈单层整齐排列,外周血Th17比例、Th17/Treg降 低,Treg比例升高,异位内膜组织IL-17A、RORγt mRNA和 蛋 白 水 平 降 低,Foxp3、GITR、CTLA-4 mRNA和蛋白水平升高,血清IL-6、IL-17A、IL-22水平降低,IL-10、TGF-β水平升高(P<0.05);且HXZS-H组效果优于HXZS-L组(P<0.05).结论:活血滋肾方能够调节EMs大鼠Th17/Treg及其相关炎症因子表达平衡,并抑制异位病灶生长.
Objective:To explore the effect of Huoxue Zishen Decoction on helper T cell 17/regulatory T cell(Th17/Treg)bal-ance and the expression of inflammatory factors in endometriosis(EMs)rats.Methods:A total of 60 rats were randomly divided into Sham group,EMs group,low-dose Huoxue Zishen Decoction(HXZS-L)group,and high-dose Huoxue Zishen Decoction(HXZS-H)group,with 15 rats in each group.Rat models of EMs were established by autologous loendometrial transplantation.Rats in Sham group only underwent ligation and resection of the left uterine horn,without transplantation.After 2 weeks of modeling,rats in HXZS-L group and HXZS-H group were orally administered with Huoxue Zishen Decoction(10.71 g/kg,42.84 g/kg)daily;rats in Sham group and EMs group were orally administered an equal amount of physiological saline for 3 consecutive weeks.After 3 weeks of treat-ment,ELISA was used to detect inflammatory factors IL-6,IL-17A,IL-22,IL-10 and TGF-β levels in serum;HE staining was used to observe ectopic endometrial tissue damage;flow cytometry was used to detect Th17/Treg in peripheral blood;RT-qPCR,Western blot were used to detect Th17/Treg related markers(IL-17A,RORγt,Foxp3,GITR,CTLA-4)mRNA and protein levels in ectopic endometrial tissue.Results:Compared with Sham group,the endometrium lesions of rats in EMs group were raised and transparent,with a large number of blood vessels covering the surface,a large number of blood vessels were formed around,and ectopic endometrial epithelial cells were arranged in a multi-layered disordered manner,the proportion of Th17 and Th17/Treg in peripheral blood were in-creased,the proportion of Treg was decreased,and the IL-17A,RORγt mRNA and protein levels in ectopic endometrial tissue were increased,while Foxp3,GITR,CTLA-4 mRNA and protein levels were decreased,IL-6,IL-17A,and IL-22 levels in serum were in-creased,IL-10,TGF-β levels in serum were decreased(P<0.05);compared with EMs group,the ectopic endometrial lesions of rats in HXZS-L group and HXZS-H group were reduced,with a small number of blood vessels were formed around,and ectopic endometrial epithelial cells were arranged in a single layer in an orderly manner,the proportion of Th17 and Th17/Treg in peripheral blood were de-creased,the proportion of Treg was increased,and the IL-17A,RORγt mRNA and protein levels in ectopic endometrial tissue were decreased,while Foxp3,GITR,CTLA-4 mRNA and protein levels were increased,IL-6,IL-17A and IL-22 levels in serum were de-creased,IL-10,TGF-β levels in serum were increased(P<0.05);and the effect of HXZS-H group was better than that of HXZS-L group(P<0.05).Conclusion:Huoxue Zishen Decoction could regulate balance of Th17/Treg and the expression of related inflammato-ry factors in EMs rats,and inhibit the growth of ectopic lesions.
