硫代牛磺酸抵御皮肤光损伤的作用机制研究OA
Research on the Mechanisms of Thiotaurine in Protecting Against Photodamage
目的:探究硫代牛磺酸抵御皮肤光损伤的修护机理.方法:建立光诱导损伤模型角质细胞及3D表皮模型,检测光损伤后的细胞活力和活性氧(Reactive Oxygen Species,ROS),光损伤3D表皮模型的组织活力、丝聚蛋白(Filaggrin,FLG)、兜甲蛋白(Loncrin,LOR)和核纤层蛋白B(Lamin-B)的免疫荧光结果及炎症因子.结果:通过光诱导角质细胞模型可知,硫代牛磺酸能提升光损伤后细胞活力,且能降低ROS产生;光诱导3D表皮模型通过增加FLG、LOR的表达修护表皮屏障,增加细胞核衰老标志物Lamin-B表达抵御细胞衰老,且能降低白细胞介素-1α(Interleukin-1 alpha,IL-1α)分泌降低炎症.结论:硫代牛磺酸能保护皮肤免受光诱导的损伤.
Objective To explore the repair mechanism of thiotaurine in resisting skin photodamage.Methods A model of light-induced damage was established using keratinocytes and a 3D epidermal model.The study assessed cell viability and Reactive Oxygen Species(ROS)levels post-light damage,as well as tissue vitality,immunofluorescence results for Filaggrin(FLG),Loricrin(LOR),and Lamin-B in the 3D epidermal model,and inflammatory factors.Results It can be known from the light-induced keratinocyte model that thiotaurine can enhance the cell viability after light damage and reduce the production of ROS.The light-induced 3D epidermal model repairs the epidermal barrier by increasing the expression of FLG and LOR,increases the expression of the nuclear senescence marker Lamin-B to resist cellular senescence,and can reduce the secretion of Interleukin-1 alpha(IL-1α)to reduce inflammation.Conclusion Thiotaurine can protect the skin from light-induced damage.
