目的:探究阿司匹林对瘢痕疙瘩的抑制作用,评估其作为临床备选治疗药物的潜力.方法:本实验通过组织块法获得瘢痕疙瘩来源的原代成纤维细胞,经阿司匹林处理后收集细胞进行转录组测序,通过CCK-8法检测阿司匹林对细胞增殖水平的影响,流式细胞术检测细胞周期及凋亡情况,并采用基质胶构建人源性组织异种移植模型,验证阿司匹林处理后的细胞成瘤能力.结果:转录组测序分析表明,阿司匹林处理组与对照组相比共检测出5 849个差异基因,差异基因功能主要集中于细胞周期、DNA复制、p53信号通路等,均直接影响细胞增殖、凋亡水平;细胞增殖、周期及凋亡检测证实阿司匹林抑制瘢痕疙瘩来源成纤维细胞增殖并诱导细胞凋亡,并通过停滞细胞周期抑制细胞增殖;人源性组织异种移植模型表明阿司匹林处理后细胞成瘤体积显著变小.结论:本研究结果表明,阿司匹林可以抑制瘢痕疙瘩来源的成纤维细胞增殖、促进凋亡,裸鼠在体实验也表明阿司匹林处理后细胞成瘤体积显著减少,因此,提示阿司匹林有望作为瘢痕疙瘩的潜在治疗药物.

Objective As a benign fibroproliferative tumor,keloid is extremely prone to recurrence in the clinic regardless of surgical excision or drug injection,and there is no good treatment.Therefore,exploring and expanding more effective treatment modalities or drugs is an urgent clinical problem.Aspirin is a commonly used drug in the clinical treatment of cardiovascular and cerebrovascular diseases,and it has an antifibrotic effect,therefore,this paper initially explores whether it has an inhibitory effect on keloid scars,and whether it can be used as an alternative drug in the clinical treatment of keloid scars.Methods In this experiment,primary fibroblasts of keloid origin were obtained by the tissue block method,and the cells were collected for transcriptome sequencing after aspirin treatment,the effect of aspirin on the proliferation level of the cells was detected by the CCK-8 assay,and the cell cycle and apoptosis were detected by flow cytometry,and human tissue-derived xenograft models of keloid was constructed by using matrix gel to validate the tumorigenic ability of the cells after aspirin treatment.Results Transcriptome sequencing analysis showed that a total of 5 849 differential genes were detected in the aspirin-treated group compared with the control group,and the functions of the differential genes were mainly focused on the cell cycle,DNA replication,and the p53 signaling pathway,which all had a direct influence on the levels of cell proliferation and apoptosis;cell proliferation,cell cycle,and apoptosis assays demonstrated that aspirin inhibited the proliferation of fibroblasts of keloidal origin,induced apoptosis,and inhibited cell proliferation through cell cycle arrest;human tissue-derived xenograft models showed that the size of the xenografts became significantly smaller after aspirin treatment.Cell proliferation,cell cycle and apoptosis assays confirmed that aspirin inhibited the proliferation of keloid-derived fibroblasts and induced cell apoptosis,and inhibited cell proliferation through cell cycle arrest.Conclusion The present study demonstrates that aspirin can inhibit the proliferation and promote apoptosis of keloid-derived fibroblasts,and in vivo experiments in nude mice also demonstrates a significant reduction in the tumorigenic volume of the xenografts after treatment with aspirin,thus suggesting that aspirin is promising as a potential therapeutic agent for keloid scars.
