响应面试验优化黑皮鸡枞液体菌种培养条件OA
Response Surface Test to Optimise the Culture Conditions of Liquid Strains of Oudemansiella raphanipes
黑皮鸡枞作为近年来成功商品化的新晋食用菌,富含多种营养物质,但是目前对其液体菌种培养条件的优化研究还不够深入,未能使其达到最佳效益.研究通过采用单因素筛选,以黑皮鸡枞菌丝体生物量为响应值,获得最佳的培养温度、接种量、摇床转速、培养时间,利用 Box-Behnken 中心复合试验分析得到最优培养条件并验证.结果表明:黑皮鸡枞最佳培养条件为培养温度 26℃,接种量 11%,摇床转速 145 r/min,培养时间 10 d,菌丝体生物量可达到(1.213±0.022)g/100 mL,与理论值结果相拟合,表明优化的黑皮鸡枞液体菌种培养条件合理,研究可为黑皮鸡枞液体菌种高效繁育及规模化栽培奠定基础.
As a newly emerging edible fungus successfully commercialised in recent years,Oudemansiella raphanipes is rich in a variety of nutrients,but the current research on the optimisation of the culture conditions of its liquid strains has not been thorough enough to achieve the optimal benefits.In this study,the optimal culture temperature、inoculum amount、shaker speed and incubation time were obtained by using one-way screening with the mycelial bi-omass of Oudemansiella raphanipes as the response value,and the optimal culture conditions were obtained and ver-ified by using Box-Behnken central composite test analysis.The results showed that the optimal culture conditions of Oudemansiella raphanipes were culture temperature 26℃,inoculum amount 11%,shaker speed 145 r/min and culture time 10 d.The biomass of mycelium could reach(1.213±0.022)g/100 mL,which was fitted with the theoret-ical value,indicating that the optimized culture conditions of Oudemansiella raphanipes liquid strain were reasona-ble.The study can lay the foundation for the efficient breeding and large-scale cultivation of the liquid strain.
