质谱技术在临床药物体内定量分析中的应用与精准医疗实践进展OA
Applications of mass spectrometry technology in clinical drug quantitative analysis and precision medicine practices
以治疗药物监测为代表的临床药物体内定量分析是精准医疗的关键环节之一.质谱技术凭借其高灵敏度、高分辨率及多组分分析能力,逐步代替传统定量技术或在特定场景下作为其有效补充,可共同提升检测方法的精确性与效率,成为临床药物定量分析的核心工具.近年来,质谱技术得到了快速发展,除传统的三重四极杆质谱外,还有飞行时间质谱、轨道阱质谱、电感耦合等离子体质谱、基质辅助激光解吸电离质谱、直接实时分析质谱和探针电喷雾电离源质谱等新型质谱技术,这些质谱技术融合了高通量、高特异性与实时监测能力,已成为精准医疗实践的重要支撑技术.通过对上述6类质谱技术的原理及应用进行综述,以期推动治疗药物监测从经验驱动转向数据驱动,为个体化医疗提供了技术支持.
The process of clinical pharmaceutical quantitative analysis,represented by therapeutic drug monitoring(TDM),serves as a key component of precision medicine.Leveraging high sensitivity,high resolution,and multi-component analysis capabilities,mass spectrometry(MS)is gradually replacing traditional quantitative techniques or serving as an effective supplement to them in specific scenarios,significantly enhancing the accuracy and efficiency of TDM,and emerging as a core tool in clinical pharmaceutical quantitative analysis.In recent years,mass spectrometry technology has undergone rapid development.Beyond traditional triple quadrupole mass spectrometer(QQQ-MS),novel mass spectrometer techniques such as time-of-flight mass spectrometer(TOF-MS),orbitrap mass spectrometer(Orbitrap-MS),inductively coupled plasma mass spectrometer(ICP-MS),matrix-assisted laser desorption/ionization mass spectrometer(MALDI-MS),direct analysis in real-time mass spectrometer(DART-MS),and probe electrospray ionization mass spectrometer(pESI-MS)have emerged.These technologies integrate high throughput,high specificity,and real-time monitoring capabilities,and have become essential supporting technologies in precision medical practice.This review focuses on the principles and applications of the above six types of mass spectrometry technologies,aiming to advance TDM from an experience-driven approach to a data-driven one,thereby providing technical support for personalized medicine.
