用微量检测联合HPLC-MS/MS法测定癫痫儿童血浆中唑尼沙胺浓度OA
Determination of zonisamide in epilepsy children by micro-detection technology combined with HPLC-MS/MS
目的 建立微量检测技术结合液相色谱-质谱联用法(HPLC-MS/MS)测定癫痫儿童血浆中唑尼沙胺的浓度.方法 取血浆样本10 μL经有机溶剂沉淀蛋白进行前处理,采用反相色谱柱,流动相:0.075%甲酸-2 mmoL·L-1乙酸铵-水(A)和甲醇(B)进行梯度洗脱,流速为0.45 mL·min-1,柱温为50℃,进样量为1 μL.定量方法采用内标法,内标为唑尼沙胺-15N-D4.采用正离子电喷雾离子源,多反应监测模式扫描.考察该方法的专属性、标准曲线、定量下限、准确度和精密度、残留效应、提取回收率、基质效应和稳定性.结果 唑尼沙胺和内标的保留时间均为1.25 min,在其保留时间内无内源性物质干扰.血浆样品中唑尼沙胺在1~62.5 μg·mL-1线性范围内关系良好(r=0.999 7),定量下限为1 μg·mL-1.批内和批间准确度为97.75%~107.95%,准确度标准误均≤7.95%,精密度相对标准偏差均≤4.84%,不受血浆基质的影响,稳定性良好,均符合ICHM10《生物分析方法验证指导原则》要求.结论 本方法微量采血、操作简便、高效快速,可用于临床癫痫患儿的唑尼沙胺日常治疗药物监测.
Objective To establish a method for determination of zonisamide in plasma of epileptic children by micro-detection technology combined with high performance liquid chromatography-mass spectrometry(HPLC-MS/MS).Methods 10 μL of plasma samples were pre-treated by precipitation protein method with organic solvent.Reversed-phase chromatographic columns were adopted.Flow phase:0.075%formic acid-2 mmol·L-1 ammonium acetate-water(A)and methanol(B),gradient elution;flow speed:0.45 mL·min-1;injection volume:1 μL.The quantitative method adopted the internal standard method,and the isotope internal standard was zonisamide-15N-D4.Ion source was electric spray ion(ESI)source,multireaction monitoring(MRM)mode for quantitative analysis,using positive ion monitoring mode.The specificity,standard curve,lower limit of quantitation(LLOQ),residual effect,precision,matrix effects,extraction recovery and stability were investigated.Results The retention times of zonisamide and its internal standard were both 1.25 min,and there was no interference from endogenous substances within the retention time.The linear range of zonisamide was 1-62.5 μg·mL-1(r=0.999 7),and LLOQ was 1 μg·mL-1.The intra-day and inter-day precision RSD was equal or less than 4.84%,and the accuracy ranges from 97.75%-107.95%,with good stability,and this method was not affected by common matrix.Conclusion This method involves micro-blood collection,which is simple to operate,highly efficient and rapid,and can be used for the monitoring of daily therapeutic drugs in children with epilepsy in clinical practice.
