一株丹顶鹤源鼠鼻罗氏菌的致病性及耐药性分析OA
Pathogenicity and Antibiotic Resistance of a Strain of Rothia nasimurium Derived from Red-crowned Crane
为确定江苏省盐城国家级珍禽自然保护区内丹顶鹤发病的原因并制定合理治疗措施,采集发病丹顶鹤口咽及肛拭子样品,进行病原分离鉴定及致病性和耐药性分析.结果显示:分离获得一株革兰氏阳性菌;该菌在鲜血培养基上形成光滑、淡灰色、圆形菌落;经生化鉴定,葡萄糖、枸橼酸盐和氨基酸代谢阳性;用DNA提取试剂盒提取菌液DNA,通过16S rDNA PCR扩增测序确认为鼠鼻罗氏菌.对1周龄雏鸭通过口服感染进行致病性试验,攻毒1 d后雏鸭出现剂量依赖性脱毛症状,3 d后到达高峰,之后逐渐恢复;剖检可见特征性肺部充血出血,其他脏器未见明显病变.使用抗菌药物敏感试验纸片法进行药敏试验,发现该菌对氨基糖苷类部分抗生素具有耐药性,对其他类抗生素敏感.本研究在丹顶鹤群体中检出致病性鼠鼻罗氏菌,并分析了其耐药情况,为珍稀鸟类病原监测及保护区疾病防控提供了依据.
In order to identity morbidity of red-crowned cranes in Yancheng National Rare Birds Nature Reserve in Jiangsu Province,and to develop reasonable countermeasures,oropharyngeal and cloacal swab samples were collected from affected cranes for pathogen isolation and identification,as well as pathogenicity and antibiotic resistance analysis.The results showed that a Gram-positive bacterium was isolated,which formed smooth,light gray,circular colonies on blood agar;positive reactions for glucose,citrate and amino acid metabolism were determined by biochemical identification tubes;bacterial DNA was extracted using a DNA extraction kit,and confirmed as Rothia nasimurium through 16S rDNA PCR amplification and sequencing.One-week-old ducklings were infected via oral for pathogenicity test,the symptoms of dose-dependent feather loss appeared one day post-infection,peaking after three days,and gradually recovering thereafter;characteristic pulmonary congestion and hemorrhage were revealed by post-mortem examination,but no significant lesions were observed in other organs.Disk diffusion antibiotic susceptibility test revealed that the strain was resistant to some aminoglycoside antibiotics,but sensitive to other classes of antibiotics.Pathogenic Rothia nasimurium was detected from a red-crowned crane population,and analyzed for its antibiotic resistance,providing a basis for pathogen monitoring of rare birds and disease prevention in the reserve.
