分泌关联的Ras相关GTP酶1B(secretion-associated Ras-related GTPase 1B,SAR1B)的经典功能表现为在包被蛋白复合体II囊泡形成与出芽中的核心驱动作用,以及对蛋白质与脂质运输的调控.近年研究表明,SAR1B亦可作为亮氨酸传感器,通过与GATOR2(GTPase-activating protein activity toward Rags 2)复合物的亮氨酸依赖性相互作用调控哺乳动物雷帕霉素靶蛋白复合物1信号通路,影响蛋白质合成.在病理方面,SAR1B突变可致常染色体隐性遗传病即乳糜微粒潴留病,引发脂肪吸收不良等症状;且可影响肝脏与肺肿瘤发展以及肿瘤细胞耐受性.本文综述了SAR1B促成囊泡形成与出芽的机制,以及在多种病理生理过程中SAR1B所发挥的作用.

The canonical role of secretion-associated Ras-related GTPase 1B(SAR1B)involves the critical regulation of coat protein complex II vesicle formation and budding,as well as the modulation of protein and lipid trans-port.Recent studies have demonstrated that SAR1B also functions as a leucine sensor,modulating the mammalian target of rapamycin complex 1 signaling pathway through leucine-dependent interaction with GTPase-activating protein activity to-ward Rags 2 complex,thereby affecting protein synthesis.In pathological contexts,mutations in SAR1B cause an autoso-mal recessive disorder known as chylomicron retention disease,characterized by fat malabsorption and associated clinical manifestations.Additionally,SAR1B has been implicated in the progression of hepatic and pulmonary tumors and in sus-taining cancer cell survival.This review summarizes the mechanism by which SAR1B promotes vesicle formation and bud-ding,and its multifaceted roles in diverse pathophysiological processes.
