心血管疾病(cardiovascular diseases,CVD)作为全球首要致死病因,其病理机制复杂,涉及线粒体功能障碍(如能量代谢异常、氧化应激、炎症反应等)的多个方面.研究显示,蛋白质乙酰化修饰在调控线粒体功能方面发挥核心作用.沉默信息调节因子3(silent information regulator 3,SIRT3)作为线粒体基质中活性最高的脱乙酰酶,通过调控关键蛋白的去乙酰化修饰,维持线粒体能量代谢平衡、氧化还原稳态及结构与功能完整性.本文综述了SIRT3介导线粒体结构与功能的作用机制,及其在高血压、动脉粥样硬化、动脉瘤、心肌肥厚等多种CVD病理进程中的调控作用,并展望了SIRT3作为潜在治疗靶点的应用前景,旨在为CVD的临床治疗提供参考资料.

Cardiovascular diseases(CVD)remain the leading cause of mortality worldwide.Their complex pathogenesis is closely associated with mitochondrial dysfunction,including impaired energy metabolism,excessive oxida-tive stress,and dysregulated inflammatory responses.Increasing evidence indicates that protein acetylation is a critical post-translational modification governing mitochondrial function.Silent information regulator 3(SIRT3),the most active nicotinamide adenine dinucleotide-dependent deacetylase within the mitochondrial matrix,plays a pivotal role in maintain-ing mitochondrial homeostasis.By deacetylating multiple key mitochondrial proteins,SIRT3 regulates energy metabolism,preserves redox balance,and sustains mitochondrial structural and functional integrity.This review summarizes the molec-ular mechanisms by which SIRT3 modulates mitochondrial structure and function and highlights its involvement in the pathogenesis of major CVD,including hypertension,atherosclerosis,aneurysms,and cardiac hypertrophy.Finally,we discuss the therapeutic potential of targeting SIRT3,providing insights that may facilitate the development of novel strate-gies for the clinical management of cardiovascular diseases.
