响应面法优化大孔树脂纯化毛连菜中有机酸的工艺研究OA
Optimization of the Purification Process for Organic Acids from Picris hieracioides L.Using Macroporous Resin by Response Surface Methodology
目的:基于单因素实验结果,采用Box-Behnken响应面法优化大孔树脂分离纯化毛连菜中有机酸的工艺.方法:以低共熔溶剂提取的毛连菜中绿原酸等有机酸为对象,通过静态吸附-解吸附实验比较7种大孔树脂的吸附与解吸附性能,筛选最优树脂;采用单因素实验考察高径比、洗脱溶剂浓度和体积对纯化效果的影响,并在此基础上通过响应面法优化上述三个关键参数,确定最佳纯化工艺.结果:确定采用NKA-2型大孔树脂为纯化有机酸的最优树脂,最佳纯化条件为高径比8∶1,洗脱溶剂乙醇浓度80%,洗脱体积3BV.在该条件下,毛连菜有机酸的总回收率达到90.38%,与预测值的相对误差为0.64%.结论:NKA-2树脂纯化毛连菜有机酸的工艺效果较好,优化的工艺稳定可行,具备一定的工业应用潜力.
Objective To optimize the purification process for organic acids from Picris hieracioides L.using macroporous resin by the Box-Behnken response surface methodology based on single-factor experiments.Methods Organic acids(pri-marily chlorogenic acid)extracted from P.hieracioides using deep eutectic solvents were selected.The optimal macroporous resin was screened by comparing the adsorption and desorption capacities of seven resins through static experiments.The effects of aspect ratio,eluent concentration,and elution volume on the purification were investigated using single-factor ex-periments.Subsequently,response surface methodology was employed to optimize these three critical parameters to deter-mine the optimal purification conditions.Results The NKA-2 macroporous resin was identified as the most suitable for pu-rifying organic acids.The optimal purification conditions were an aspect ratio of 8∶1,an eluent concentration of 80%,and an elution volume of 3 BV.Under these conditions,the total recovery rate of organic acids from P.hieracioides reached 90.38%,with a relative error of 0.64%compared to the predicted value.Conclusion The NKA-2 resin demonstrated ex-cellent performance in purifying organic acids from P.Hieracioides.The optimized process is stable and feasible,showing potential for industrial application.
