低共熔溶剂提取绞股蓝黄酮类成分的工艺优选OA
Optimization of Deep Eutectic Solvent Extraction Process of Flavonoids from Gynostemma pentaphyllum
目的 优选绞股蓝中芦丁、槲皮素、山柰素等 3 种黄酮类成分的提取工艺.方法 采用高效液相色谱(HPLC)法测定含量,以绞股蓝中芦丁、槲皮素、山柰素 3 种黄酮类成分的总提取率为评价指标,考察低共熔溶剂的类型、摩尔比、含水量,确定最佳低共熔体系,结合Box-Behnken响应曲面法考察提取液料比、超声温度、超声时间对提取工艺的影响,确定最佳提取工艺.结果 绞股蓝黄酮类成分低共熔溶剂提取的最佳工艺为:摩尔比 3∶1,含水量 30%的氯化胆碱/草酸为提取溶剂,液料比 35 mL·g-1,超声温度 69℃,超声时间 40 min.结论 优选的绞股蓝低共熔溶剂提取工艺稳定可行,可为绞股蓝黄酮类成分的高效、安全提取提供参考.
Objective To optimize the extraction process of rutin,quercetin,and kaempferol,three major flavonoid components,from Gynostemma pentaphyllum.Methods A HPLC method was used to analyze the content.The total extraction yield of three flavonoid components-rutin,quercetin,and kaempferol-was selected as the evaluation index.The optimal deep eutec-tic solvent system was determined by examining the type,molar ratio,and water content of the deep eutectic solvent.Box-Behnken response surface methodology was used to investigate the effects of liquid-solid ratio,ultrasonic temperature,and ultrasonic time on the extraction of Gynostemma pentaphyllum flavonoid components to determine the optimal extraction conditions.Results The optimum extraction process of three flavonoid components from Gynostemma pentaphyllum was as follows:the solvent composition consists of choline chloride and oxalic acid at a molar ratio of 3∶1,with a water content of 30%.Additionally,the liquid-to-solid ratio is maintained at 35 mL·g-1,the ultrasonic temperature is set at 69℃,and the ultrasonic duration is 40 minutes.Conclu-sion The optimized deep eutectic solvent-based extraction method demonstrates stability and feasibility,providing a reliable ap-proach for the efficient and safe extraction of flavonoids from Gynostemma pentaphyllum.
