Advances in the Anti-Inflammatory Effects of Dichloroacetate in Inflammatory Diseases
二氯乙酸盐是丙酮酸脱氢酶激酶(PDK)的泛抑制剂.PDK 抑制丙酮酸脱氢酶复合物(PDC),使细胞代谢从氧化磷酸化转向有氧糖酵解(即Warburg效应).二氯乙酸盐通过抑制 PDK,激活 PDC,可逆转有氧糖酵解,增强线粒体氧化磷酸化并减少乳酸产生.近年来,随着糖酵解通路在肿瘤及炎症性疾病研究中的深入,越来越多的研究发现二氯乙酸盐在炎症性疾病中的潜在应用价值.该文综述二氯乙酸盐在各种炎症性疾病中的抗炎活性及其机制研究进展,为二氯乙酸盐在炎症性疾病中基础研究和临床应用提供参考,并为炎症性疾病的治疗提供新的视角.
Dichloroacetate(DCA)is a pan-inhibitor of pyruvate dehydrogenase kinase(PDK).PDK inhibits the pyru-vate dehydrogenase complex(PDC),thereby promoting a shift in cellular metabolism from oxidative phosphorylation towards aero-bic glycolysis(the Warburg effect).By inhibiting PDK,DCA activates PDC,reverses aerobic glycolysis,enhances mitochondrial oxidative phosphorylation,and reduces lactate production.In recent years,with the in-depth study of the glycolytic pathway in tumors and inflammatory diseases,an increasing number of studies have revealed the potential clinical value of DCA in inflamma-tory diseases.This article reviews the research progress regarding the anti-inflammatory activity and mechanisms of DCA in various inflammatory diseases,aiming to provide references for both basic research and clinical applications of DCA in these conditions,as well as to offer new perspectives for the treatment of inflammatory diseases.
