目的 探究结直肠癌荷瘤小鼠化疗期间使用益生元和益生菌干预对小鼠肠道微生物群结构的影响.方法 将CT26 细胞经皮下注射到BALB/c小鼠,构建结直肠癌荷瘤小鼠模型,将小鼠分组为对照组(生理盐水 0.5 ml/d)、化疗(HS)组(20 mg/kg 5-FU)、益生菌(HYSJ)组(双歧杆菌三联活菌)、益生元(HYSY)组(低聚果糖增强菊粉)、益生菌+益生元(HYSJYSY)组(双歧杆菌三联活菌+低聚果糖增强菊粉),上述药物持续灌胃 3 周后,处死动物取肿瘤组织,分别评价肿瘤体积和质量,并取各组小鼠粪便进行肠道 16S rRNA V3 和V4 可变区基因测序.结果 与对照组相比,HS组、HYSJ组、HYSY组、HYSJYSY组小鼠肿瘤质量有所降低,但差异不显著(P>0.05);与HS组相比,HYSJ组、HYSY组、HYSJYSY组体积降低(P<0.05),HYSJYSY组对细菌丰富度和均匀性有影响,并对肠道微生物群的组成有显著影响.5-FU治疗后,分析模型小鼠粪便样本中微生物变化,结果表明与HS组相比,HYSJYSY组中厚壁菌(Firmicutes)、拟杆菌(Bacteroidetes)、原细菌(Proteobacteria)、梭杆菌(Fusobacteria)富集水平较高,而铁杆菌(Tenericutes)、放线菌(Actinobacteria)、糖化菌(TM7)、疣状菌(Verrucomicrobia)富集水平较低(P<0.05).结论 益生元联合益生菌在 5-FU治疗过程中,可增强化疗效果,具有抗肿瘤作用,改善肠道微生物结构.

Objective To investigate the effect of prebiotic and probiotic on the structure of intestinal microbiota in colorectal cancer tumor-bearing mice during chemotherapy.Methods CT26 cells were subcutaneously injected into BALB/c mice to construct a tumor-bearing mice model of colorectal cancer.The mice were divided into control group(normal saline 0.5 ml/d),chemotherapy group(HS)group(20 mg/kg 5-FU),probiotic(HYSJ)group(bifidobacterium triple viable),prebiotic(HYSY)group(fructo-oligosaccharides enhance inulin),probiotic+prebiotic(HYSJYSY)group(bifidobacterium triple viable+fructo-oligosaccharides enhance inulin).After 3 weeks of continuous gavage of the above drugs,the animals were sacrificed and the tumor tissues were taken to evaluate the tumor volume and quality.The feces of each group of mice were taken for intestinal 16S rRNA V3 and V4 variable region gene sequencing.Results Compared with the control group,the tumor weight of mice in HS group,HYSJ group,HYSY group and HYSJYSY group was decreased,but the difference was not significant(P>0.05).Compared with HS group,the volume of HYSJ group,HYSY group and HYSJYSY group was decreased(P<0.05).HYSJYSY group had an effect on bacterial richness and uniformity,and had a significant effect on the composition of intestinal microflora.After 5-FU treatment,the microbial changes in the fecal samples of the model mice were analyzed.The results showed that compared with the HS group,the enrichment levels of Firmicutes,Bacteroidetes,Proteobacteriaand Fusobacteriain the HYSJYSY group were higher,while the enrichment levels of Tenericutes,Actinobacteria,TM7 and Verrucomicrobia were lower(P<0.05).Conclusion Prebiotic combined with probiotic can enhance the effect of chemotherapy,have anti-tumor effect and improve the intestinal microbial structure during 5-FU treatment period.
