Research Progress on the Relationship Between Ras-Raf-MEK1/2-ERK1/2 Signaling Pathway and Auditory Degeneration
丝裂原活化蛋白激酶(MAPKs)信号通路在生物信号传递中发挥着至关重要的作用.在正常和病理条件下,这些途径激活相应的下游信号因子,参与细胞增殖、分化和凋亡,以及与氧化应激相关的损伤和修复过程.MAPK家 族 中,c-Jun氨基末端激酶(JNK)与多种生物细胞的凋亡密切相关,而细胞外信号调节激酶(ERK)是该家族的另一个重要成员,与JNK不同,ERK及其相关信号通路分子(Ras-Raf-MEK1/2-ERK1/2)具有抑制细胞凋亡和调节细胞生长、发育和分裂的能力.目前,多项研究发现通过调控MAPK通路中ERK的磷酸化过程,可改善神经元功能障碍及退化等病理现象,而Ras-Raf-MEK1/2-ERK1/2 信号通路在年龄相关性听力损失方面的研究相对较少.听觉退行性病变是造成年龄相关性听力下降的一个重要原因,本文主要对Ras-Raf-MEK1/2-ERK1/2 通路与听神经退行性病变的相关性进行综述,以期为年龄相关性听力障碍的病因及治疗提供一定的理论依据.
Mitogen-activated protein kinase(MAPK)signaling pathways play a crucial role in information transmission in animals.Under both normal and pathological conditions,these pathways activate corresponding downstream signaling factors that participate in cell proliferation,differentiation,and apoptosis,as well as in the damage and repair processes associated with oxidative stress.Among the MAPK family,c-Jun N-terminal kinase(JNK)is closely linked to the apoptosis of various biological cells,while extracellular signal-regulated kinase(ERK)represents another significant member of this family.Unlike JNK,ERK and its associated signaling pathway molecules(Ras-Raf-MEK1/2-ERK1/2)possess the ability to inhibit apoptosis and regulate cell growth,development,and division.Currently,numerous studies have demonstrated that pathological phenomena such as neuronal dysfunction and degeneration can be ameliorated by modulating the ERK phosphorylation process within the MAPK pathway.Additionally,the Ras-Raf-MEK1/2-ERK1/2 signaling pathway is critical in the context of age-related hearing loss.However,relatively little research has been conducted on auditory degeneration,which is a significant contributor to age-related hearing loss.This article aims to review the correlation between the Ras-Raf-MEK1/2-ERK1/2 pathway and auditory neurodegeneration,thereby providing a theoretical foundation for exploring the causes and potential treatments of age-related hearing impairment.
