罗替戈汀山嵛酸酯对帕金森抑郁症大鼠的治疗作用OA
Therapeutic Effect of Rotigotine Behenate on Parkinson's Depression Rats
旨在探讨罗替戈汀山嵛酸酯(Rotigotine Behenate,ROTB)对帕金森病抑郁(Depres-sion in Parkinson's disease,DPD)大鼠的治疗作用.采用液相色谱-串联质谱(LC-MS/MS)法分析测定大鼠血浆中罗替戈汀的浓度;采用 6-OHDA 和 CUMS 诱导建立 DPD 大鼠模型,给予5.6、11.2、22.4 mg/kg ROTB 进行治疗.通过强迫游泳、旷场和糖水偏好实验检测 ROTB 对 DPD 大鼠抑郁症状的改善作用;Western blot 实验检测大鼠前额叶皮层 BD-NF、TrkB、PI3K、Akt、p-Akt、GSK-3β、p-GSK-3β 的表达水平;ELISA 实验检测大鼠前额叶皮层 DA 和5-HT 的含量;HE 染色观察大鼠前额叶皮层神经元病理变化.结果显示,大鼠单次肌肉注射给予ROTB,Rotigotine 可持续平稳释放28 d;ROTB 可改善DPD 大鼠抑郁症状,该作用可能与上调 DA 和 5-HT 含量,增强 BDNF-TrkB 及其下游 PI3K-Akt-GSK-3β 信号通路,改善前额叶皮层神经元细胞损伤有关.以上结果表明,ROTB 对 DPD 大鼠具有一定的治疗作用.
This experiment aims to explore the therapeutic effect of Rotigotine Behenate(ROTB)on depression in Parkinson's disease(DPD)rats.Rotigotine concentrations in rat plasma were determined by liquid chromatogra-phy-tandem mass spectrometry(LC-MS/MS).DPD rats were induced using 6-OHDA and CUMS and treated with ROTB at a doses of 5.6,11.2 and 22.4 mg/kg.The antidepressant effect of ROTB in DPD rats was assessed using forced swimming,open field,and sucrose preference tests.Expression levels of BDNF,TrkB,PI3K,Akt,p-Akt,GSK-3β and p-GSK-3β were measured by Western blot,while DA and 5-HT contents in the prefrontal cortex were analyzed by ELISA.Pathological changes in neurons in the prefrontal cortex were observed using HE staining.The results showed that a single intramuscular injection of ROTB enabled continuous and stable release of rotigotine for 28 days.ROTB alleviated depressive symptoms in DPD rats,potentially by up-regulating DA and 5-HT levels,enhancing the BDNF-TrkB and PI3K-Akt-GSK-3β signaling pathways,and reducing neuronal damage in the prefrontal cortex.These findings suggest that ROTB has therapeutic potential for treating depression in DPD rats.
