通过监测藻蓝蛋白 PC645 在不同光区的吸收光谱和圆二色(CD)谱随 pH 变化情况,了解其色基和蛋白空间结构稳定性及与能量传递功能的关系.结果显示,1)PC645 在不同光谱区段的吸收峰、CD 谱峰及二级结构比例在 pH 3.5~10 时都很稳定,尤以 pH 5~7 时蛋白构象和功能状态最佳,且 pH 5 似乎是 PC645 生理功能变化的节点;2)在 pH 3~3.5 和 pH 10~11 时,PC645 二级结构出现小幅度变化,但三级和四级结构及能量吸收功能仍正常,说明其三、四级结构的维持对其捕光功能更重要,其中 525 nm 处 CD 谱峰可作为 PC645 四级结构完整性的标志;3)在 pH<3 和 pH>10 时,空间结构解体,在酸变性时二级结构更趋于E折叠化,在碱变性时肽链迅速趋于无规则卷曲.PC645 结构和功能对环境 pH 变化呈现高度适应性,可望开发为能够耐受不同试剂环境的荧光探针而广泛应用于多种荧光检测分析等.

By monitoring absorption spectra and circular dichroism(CD)spectra across different pH conditions,we investigated the stability of phycocyanin PC645's chromophore and spatial structure,along with its energy transfer function.The results demonstrated that:1)PC645 maintained stable absorption peaks,CD peaks,and secondary structure ratios with pH 3.5-10,with optimal protein conformation and functional state observed at pH 5-7.Nota-bly,pH 5 appeared to represent a critical transition point for physiological functional changes;2)Moderate struc-tural alterations occurred at extreme pH values(pH 3-3.5 and pH 10-11),where secondary structure changes were observed while tertiary and quaternary structures and energy absorption functions remained intact,suggesting the latter play a more crucial role in light harvesting.The 525 nm CD peak emerged as a reliable indicator of qua-ternary structural integrity;3)Significant structural disintegration occurred at pH<3 and pH>10,with acid denat-uration inducing secondary structure folding while alkaline conditions caused irregular peptide chain coiling.These findings highlight PC645's remarkable pH adaptability,suggesting its strong potential as a versatile fluorescent probe capable of withstanding diverse chemical environments for various fluorescence detection applications.
