菟丝子酊的提取工艺优化OA
Optimization of the Extraction Process of Cuscutae Semen Tincture
目的:优化菟丝子酊的提取工艺.方法:以金丝桃苷、异槲皮素、紫云英苷、山柰酚的含量(mg/g)为评价指标,利用熵权法确定权重系数,计算多指标综合评分.在单因素试验的基础上,通过Box-Behnken响应面法考察浸泡时间(天)、料液比(g/mL)、乙醇浓度(%)的影响,优选菟丝子酊的提取工艺.结果:菟丝子酊最优提取工艺为菟丝子细粉用90%乙醇按料液比1∶5(g/mL)浸泡7天;工艺验证结果显示金丝桃苷、异槲皮苷、紫云英苷、山柰酚苷的平均提取含量分别为2.306 6、0.474 7、0.587 7、0.148 1mg/g,综合评分为98.64.结论:优化的工艺稳定可行,可为菟丝子酊的提取工艺提供基础依据.
Objective:Optimize the extraction process of Cuscutae Semen Tincture.Methods:Taking the contents(mg/g)of hyperoside,isorhamnetin,astragalin,and kaempferol as evaluation indicators,the entropy weight method was used to determine the weight coefficients,and the comprehensive score of multiple indicators was calculated.Based on the single-factor test,taking the soaking time(days),the ratio of material to liquid(g/mL),and the ethanol concentration(%)as factors were investigated by the Box-Behnken response surface method to optimize the extraction process of Cuscutae Semen Tincture.Results:The optimal extraction process of Cuscutae Semen Tincture was to soak the fine powder of Cuscutae Semen in 90%ethanol at a ratio of material to liquid of 1∶5(g/mL)for 7 days.The results of process verification showed that the extracted contents(mg/g)of hyperoside,isorhamnetin,astragalin,kaempferol were 2.306 6,0.474 7,0.587 7,0.148 1 respectively,and the comprehensive score was 98.64.Conclusion:The opti-mized process is reasonable,stable and reliable,which can provide a basic basis for the extraction process of Cuscutae Semen Tincture.
