目的:优化清降和胃颗粒成型工艺,并对其吸湿性进行研究.方法:以性状、成型率、溶化性、粉体性质、口感、吸湿率等参数为评价指标,采用单因素考察和正交实验设计考察优选制剂处方和成型工艺参数,并通过连续三批小试和中试进行验证;使用 Origin软件依据本品提取物和颗粒吸湿率随时间变化的规律建立吸湿动力学模型,并确定成品的临界相对湿度(CRH).结果:制剂处方优选的填充剂为麦芽糊精,优选的矫味剂为4‰三氯蔗糖和5‰甜橙粉末香精,制剂规格为10g/袋(每g相当于饮片2.70g);制粒工艺参数优选为:压辊压力70bar,送料速度20rpm,整粒速度100rpm;本品颗粒CRH为64.28%;吸湿动力学研究结果表明本品提取物和颗粒均在Exponential模型中拟合度最好,通过拟合曲线确定在RH65%条件下本品提取物在空气中暴露时间不应超过0.66h,颗粒在空气中暴露时间不应超过3.69h,超过上述暴露时间可能会引起提取物和颗粒水分超标.结论:连续三批小试和中实验证结果表明,研究确定的制剂成型工艺稳定、可行,可指导工业化生产.

Objective:To optimize the forming process of Qingjiang Hewei Granules,and study its hygroscopicity.Meth-ods:Formulation prescription and forming process were screened with the evaluation indexes like particle characteris-tics,forming rate,solubility,powder properties,taste,moisture absorption and other parameters.The forming process was finally determined through three continuous batches of lab-scale and pilot-scale verification tests.A hygroscopic dynamical model for the humidity absorption rate of the extract and granules vs.storage time was established via Ori-gin.And the critical relative humidity(CRH)was also determined.Results:Maltodextrin is the preferred filling agent in the formulation,4‰ sucralose and 5‰ orange powder are the preferred flavoring agents,and the preparation specifi-cation is 10g/bag(equal to 27g of decoction pieces).The final granulation process parameters were determined as a roller pressure of 70 bar,a feeding speed of 20 rpm,and a collating speed of 100 rpm.The CRH of this product was de-termined as 64.28%.The results of the moisture absorption kinetics study indicated that both the extract and granules exhibited the best fit in the Exponential model.Based on the fitted curve,it was determined that the exposure time of the extract in the air at RH 65%should not exceed 0.66 hours,while the granules in the air should not exceed 3.69 hours.Exceeding the exposure times may lead the moisture content of the extract and granules exceed their limits.Conclusion:According to the verification results of three continuous batches of lab-scale and pilot-scale,the formula-tion forming process was stable and feasible,which can be used for guiding industrial production.
