Network Pharmacology-Based Investigation of the Mechanism of Action of Stroke Rehabilitation Ointment in the Treatment of Vascular Cognitive Impairment

目的:运用网络药理学方法,探讨中风康复膏治疗血管性认知障碍的作用机制,为方药使用提供理论依据.方法:首先通过TCMSP数据库筛选中风康复膏的有效成分及相应的靶点,后在 Gene-Cards、DisGeNET和 OMIM 数据库中检索"血管认知障碍",获取疾病相关靶点信息.获取药物与疾病靶点交集并制作韦恩图,利用Cytoscape软件构建蛋白质-蛋白质相互作用(PPI)网络图,筛选关键成分和核心基因.采用STRING数据库构建蛋白质-蛋白质相互作用网络,用 MCODE 插件进行聚类分析.使用 Metascape数据库核心靶点进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析.结果:筛选出中风康复膏含有效成分99种,对应靶标403个.血管性认知障碍相关的靶点1 165个,其中165个药物与疾病靶点相交.GO功能富集分析分别鉴定了5 690、493和891个与生物过程、细胞成分和分子功能相关的基因.在 KEGG 分析中,癌症通路、AGE-RAGE信号通路、MAPK信号通路是最常见的通路.结论:中风康复膏中的槲皮素、豆甾醇、木犀草素、大黄素甲醚、β-谷甾醇等成分可通过IL-6、AKT1、TNF、IL-1β、TP53等多靶点调控 MAPK、AGE-RAGE、癌症等信号通路治疗血管性认知障碍.

Objective:To explore the mechanism of vascular cognitive disorders treated by stroke rehabilitation ointment using network pharmacology,and to provide theoretical basis for the use of the formula.Methods:Firstly,the TCMSP database was used to screen the active ingredients and corresponding targets of the stroke rehabilitation ointment,and the GeneCards,DisGeNET and OMIM databases were used to search for"vascular cognitive disorders"and obtain the information of disease-related targets.The intersection of drug and disease targets was taken and a Wayne diagram was made,and a protein-protein interaction(PPI)network diagram was constructed using Cytoscape software to screen the key components and core genes,and a protein-protein interaction network was constructed in the STRING database,and then clustering analysis was carried out using the MCODE plug-in.The core targets were then analyzed for Gene Ontology(GO)function and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment using Metas-cape database.Results:The stroke rehabilitation ointment was screened to contain 99 active ingredients,corresponding to 403 targets.There were 1 165 targets related to vascular cognitive disorders,of which 165 drugs intersected with dis-ease targets.GO functional enrichment analysis identified 5 690,493 and 891 genes related to biological processes,cel-lular components and molecular functions,respectively.In KEGG analysis,cancer pathway,AGE-RAGE signaling path-way,and MAPK signaling pathway were the most common pathways.Conclusion:Quercetin,stigmasterol,lignans,rhodopsin methyl ether,and β-sitosterol in stroke rehabilitation ointment can regulate MAPK,AGE-RAGE,and cancer signaling pathways through multiple targets such as IL-6,AKT1,TNF,IL-1β,and TP53 for the treatment of vascular cognitive impairment.
