碱提罗汉果多糖乳液的制备及其保湿活性的研究OA
Preparation and moisturizing activity evaluation of alkali-extracted Siraitia grosvenorii polysaccharide emulsion
目的 旨在挖掘药食同源植物罗汉果的高值化利用潜力,通过碱提工艺制备碱提罗汉果多糖(AE-SGP),以比较AE-SGP与水提罗汉果多糖(WE-SGP)的提取得率与体外保湿活性,并以这两项指标为功效成分优化O/W型乳液配方,评价乳液的贮藏、冻融及保湿活性稳定性.方法 经乙醇脱脂、碱液回流提取,通过AB-8 大孔吸附树脂脱色、Sevag法脱蛋白等步骤纯化多糖,采用苯酚-硫酸法测定得率、称重法评价保湿活性,通过单因素试验与L9(34)正交试验优化乳液配方,并通过测试乳液的粒度及Zeta电位判断乳液的稳定性.结果 AE-SGP的提取得率为 38.96%,显著高于WE-SGP的 30.58%;AE-SGP 12 h时的体外保湿率为 49.73%,优于透明质酸(39.95%)和WE-SGP(43.73%);乳液最优配方为:水相含 0.8 g AE-SGP、92.0 mL去离子水、3.0 mL 1,2-丙二醇以及 2.0 mL低分子透明质酸,油相含 3.0 mL荷荷巴油、0.5 mL G57 乳化剂;综合评分(95.87±0.31)分;该乳液于-4℃条件下贮藏 14 d、经 3 次冻融循环后,粒径与Zeta电位无显著变化(P>0.05),无分层破乳现象,且 14 d内保湿活性稳定.结论 碱提工艺可有效提升罗汉果多糖的提取得率,且AE-SGP具备优于透明质酸的体外保湿能力,可作为天然保湿原料应用于化妆品的研发;经优化得到的AE-SGP乳液综合性能优异且稳定性良好,具备实际产业化应用的潜力.
Objective To explore the high-value utilization potential of the food-medicine homologous plant Siraitia grosvenorii,prepare alkali-extracted Siraitia grosvenorii polysaccharide(AE-SGP)through an alkaline extraction process,compare the extraction yield and in vitro moisturizing activity of AE-SGP with water-extracted Siraitia grosvenorii polysaccharide(WE-SGP),and then use AE-SGP as the functional ingredient to optimize an O/W moisturizing emulsion formula,and evaluate the storage,freeze-thaw,and moisturizing activity stability of the emulsion.Methods The polysaccharides were defatted with ethanol,extracted by alkaline reflux,and purified by decolorization with AB-8 macroporous resin and deproteinization using the Sevag method.The yield was determined by the phenol-sulfuric acid method,and moisturizing activity was evaluated by gravimetric method.The emulsion formula was optimized through single-factor experiments and an L9(34)orthogonal test,and the emulsion stability was assessed by measuring its particle size and Zeta potential.Results The yield of AE-SGP(38.96%)was significantly higher than that of WE-SGP(30.58%).The moisturizing activity of AE-SGP at 12 h was 49.73%,superior to that of hyaluronic acid(39.95%)and WE-SGP(43.73%).The optimal emulsion formula consisted of an aqueous phase containing 0.8 g AE-SGP,92.0 mL deionized water,3.0 mL 1,2-propanediol and 2.0 mL low-molecular-weight hyaluronic acid,and an oil phase containing 3.0 mL jojoba oil and 0.5 mL G57 emulsifier.The comprehensive score was 95.87±0.31.After storage at-4℃for 14 days and three freeze-thaw cycles,the emulsion showed no significant changes in particle size and Zeta potential(P>0.05),no delamination or demulsification,and its moisturizing activity remained stable within 14 days.Conclusion The alkaline extraction process can effectively increase the extraction yield of Siraitia grosvenorii polysaccharide,and AE-SGP exhibits superior in vitro moisturizing ability to hyaluronic acid,making it a natural moisturizing ingredient for cosmetic research and development.The optimized AE-SGP moisturizing emulsion demonstrates excellent comprehensive performance and good stability,showing promise for practical industrial application.
