目的:探索广佛手挥发油最佳酶辅助水蒸气蒸馏提取工艺,评估其抗氧化活性,并探究其抗氧化关键活性成分与核心靶点.方法:先通过单因素试验和响应面试验优化酶辅助水蒸气蒸馏提取广佛手挥发油的工艺,再采用气相色谱-质谱联用仪(GC-MS)进行成分检测,通过 DPPH 清除试验和磷钼络合法开展抗氧化活性评估,最后运用网络药理学和分子对接技术分析关键活性成分与核心靶点.结果:提取工艺研究结果表明,酶辅助水蒸气蒸馏法提取广佛手挥发油的最佳提取工艺参数为料液比 1g∶9mL、酶解温度为 49℃、酶添加量 0.92%、酶解时间为127 min,此条件下,广佛手挥发油的得率为 0.3901%,GC-MS 检测结果显示,从广佛手挥发油中共检测出 28 个成分,其中 D-柠檬烯和 γ-松油烯的含量最高.该提取方法下获得的广佛手挥发油无论是 DPPH·自由基清除能力测试还是磷钼酸络合的总抗氧化活性测试均表现出了良好的抗氧化活性.网络药理学和分子对接技术结果显示,广佛手挥发油抗氧化的 核 心 活 性 成 分 为 4-萜烯醇、乙酸橙花酯、薄荷酮、异丁酸芳樟酯、D-柠檬烯等,核心靶点为STAT3、ESR1、MAPK1、MAPK3、MAPK14,分子对接显示活性成分和作用靶点具有高亲和力.结论:最优工艺条件下所得的广佛手挥发油经验证具有较高的体外抗氧化活性,该挥发油可通过多成分、多通路、多靶点协同机制发挥抗氧化作用,研究可为药食同源药材广佛手的合理开发提供研究参考.

[Objective]To explore the optimal enzyme-assisted steam distillation extraction process for volatile oil from Citrus medica L.var.sar-codactylis Swingle(Guangfoshou),evaluate its antioxidant activity,and investigate the key antioxidant active components and core targets.[Methods]First,single-factor experiments and response surface methodology were used to optimize the enzyme-assisted steam distillation process for extracting volatile oil from Guangfoshou.Then,GC-MS was employed for component detection;DPPH radical scavenging assay and phospho-molybdenum complex method were applied to evaluate antioxidant activity.Finally,network pharmacology and molecular docking technology were used to analyze the key active components and core targets.[Results]The extraction process study showed that the optimal parameters for en-zyme-assisted steam distillation of Guangfoshou volatile oil were:solid-liquid ratio 1 g∶9 mL,enzymatic hydrolysis temperature 49℃,enzyme ad-dition amount 0.92%,and enzymatic hydrolysis time 127 min.Under these conditions,the yield of Guangfoshou volatile oil was 0.3901%.GC-MS detection identified 28 components in the volatile oil,among which D-limonene and γ-terpinene had the highest contents.The volatile oil ob-tained by this extraction method showed good antioxidant activity in both DPPH·radical scavenging capacity test and total antioxidant activity test via phosphomolybdenum complexation.Results from network pharmacology and molecular docking indicated that the core antioxidant active components of Guangfoshou volatile oil included 4-terpineol,neryl acetate,menthone,linalyl isobutyrate,and D-limonene,etc.,while the core targets were STAT3,ESR1,MAPK1,MAPK3,and MAPK14.Molecular docking demonstrated high affinity between the active components and the target proteins.[Conclusion]The Guangfoshou volatile oil obtained under the optimal process conditions has been verified to have high in vitro antioxidant activity.This volatile oil exerts antioxidant effects through a synergistic mechanism involving multiple components,multiple pathways,and multiple targets.This study can provide a research reference for the rational development of Guangfoshou,a medicinal and edible homologous herb.
