钙化性主动脉瓣疾病(CAVD)已成为全球范围内最常见的心脏瓣膜疾病之一,尤其在老年人群中发病率显著升高.该疾病的病理特征为主动脉瓣叶的进行性纤维化与钙盐沉积,进而导致瓣膜增厚、硬化,最终引发主动脉瓣狭窄,严重影响心脏功能并增加心血管事件风险.近年来,随着分子生物学、影像学等领域的飞速发展,其发病机制研究取得了显著进展,揭示了该疾病并非简单的退行性病变,而是一个涉及多重分子和细胞通路的主动、复杂且可调控的病理过程.文章旨在系统梳理钙化性主动脉瓣疾病在流行病学、发病机制及治疗策略方面的研究进展,重点探讨炎性反应、脂质代谢异常等关键因素在疾病发生发展中的作用,并展望当前研究的局限性与未来方向,以期为早期诊断、精准干预和新药研发提供理论依据.

Calcific aortic valve disease(CAVD)has become one of the most common heart valve diseases worldwide,with a significantly increased incidence among the elderly.The pathological features of this disease include progressive fibrosis and calcium salt deposition in the aortic valve leaflets,leading to valve thickening,hardening,and eventually aortic stenosis,which seriously affects cardiac function and increases the risk of cardiovascular events.In recent years,with the rapid develop-ment of molecular biology,imaging,and other fields,significant progress has been made in the study of its pathogenesis,re-vealing that this disease is not a simple degenerative lesion but an active,complex,and controllable pathological process invol-ving multiple molecular and cellular pathways.This review aims to systematically summarize the research progress in the epi-demiology,pathogenesis,and treatment strategies of calcific aortic valve disease,focusing on the roles of key factors such as inflammatory response and lipid metabolism abnormalities in the disease's occurrence and development,and to look forward to the limitations of current research and future directions,with the aim of providing a theoretical basis for early diagnosis,precise intervention,and new drug development.
