光敏剂BC诱导细胞凋亡抑制结直肠癌机制研究OA
Photosensitizer BC Inhibits Colorectal Cancer by Producing ROS to Induce Apoptosis
目的 探究光敏剂BC对结直肠癌细胞增殖、迁移、侵袭的影响及作用机制.方法 首先通过MTT实验评估光敏剂BC在黑暗与光照条件下对结直肠癌细胞增殖能力的抑制作用,并筛选其半数抑制浓度IC50,以此分组来进行以下实验:通过划痕以及Transwell侵袭迁移实验证明其对结直肠癌细胞侵袭迁移能力的抑制作用;通过活性氧ROS检测细胞内活性氧水平;通过线粒体膜电位检测线粒体膜电位的变化;通过Western blot实验检测结直肠癌细胞中Bcl-2和Bax蛋白的表达情况.结果 与黑暗条件相比较,光照条件下BC显著抑制结直肠癌细胞的增殖、迁移与侵袭;诱导细胞活性氧水平升高、线粒体膜电位降低;调控Bcl-2的蛋白表达水平降低、Bax的蛋白表达上升.结论 光敏剂BC在光照激活后的活性明显优于黑暗条件,并可抑制结直肠癌的增殖、侵袭和迁移能力,其可能通过产生更多ROS影响线粒体膜电位诱导结直肠癌凋亡.
Objective To investigate the effect and mechanism of photosensitizer BC on the proliferation,migration and invasion of colorectal cancer cells.Methods Firstly,the inhibitory effect of photosensitizer BC on the proliferation of colorectal cancer cells under both dark and light conditions was evaluated by an MTT assay,and its half inhibitory concentration IC50 was determined.The cells were grouped accordingly for subsequent experiments.Scratch wound healing and trans well invasion/migration assays were utilized to demonstrate the in-hibitory effects of BC on the invasive and migratory capabilities of colorectal cancer cells;intracellular reactive oxygen species(ROS)levels were measured to assess oxidative stress;changes in mitochondrial membrane potential were detected;and Western blot analysis was per-formed to examine the expression levels of Bcl-2 and Bax proteins in the colorectal cancer cells.Results Compared to dark conditions,BC significantly inhibited the proliferation,migration and invasion of colorectal cancer cells under light conditions.It induced the increase of ROS level and the decrease of mitochondrial membrane potential.Furthermore,it modulated the protein expression levels,downregulating Bcl-2 while upregulating Bax.Conclusion The activity of photodynamic compound BC after light activation is significantly better than that under dark conditions,and it can inhibit the proliferation,invasion and migration of colorectal cancer.It may induce apoptosis of colorectal cancer by generating increased levels of ROS,thereby affecting mitochondrial membrane potential.
