野生西藏沙棘多功能菌株的筛选鉴定及促生能力比较OA

以青海省野生西藏沙棘(Hippophae thibetana Schlechtend)根际土壤为材料,分离、纯化微生物菌株并进行分类鉴定,在此基础上测定菌株固氮、溶磷和解钾功能特性,筛选多重能力菌株进行促生效果评价.结果表明,共鉴定出3株假单胞菌(N7、W4、N19)、1株肠肝菌(Y65).培养5 d,4株菌株固氮透明圈直径为9.07~11.63 mm,固氮酶含量为183.41~241.38 U/L;解无机磷透明圈直径为6.80~16.43 mm,解无机磷含量为13.17~18.77 μg/mL;解钾黄色光圈直径为14.43~19.41 mm,K+含量为18.47~21.47 μg/mL.平板促生试验结果表明,4株菌株均可提高生菜种子的发芽率,发芽率为78.89%~90.00%.4株供试菌株均表现出显著的固氮、解无机磷、解钾能力和促生能力,其中Y65菌株的生物量积累效果最佳.

In this study,rhizosphere soil samples of Hippophae thibetana Schlechtend(Wild Tibetan Seabuckthorn)from Qinghai Province were collected to isolate,purify,and classify microbial strains.The functional abilities of the strains,including nitrogen fixation,phosphorus solubilization and po-tassium release,were evaluated.The growth-promoting effects of selected multifunctional strains were also assessed.The results showed that three strains of Pseudomonas(N7,W4,N19)and one strain of Enterohepater(Y65)were identified.After five days of cultivation,the diameter of the nitro-gen-fixing halo ranged from 9.07 to 11.63 mm,and nitrogenase content ranged from 183.41 to 241.38 U/L.The diameter of the phosphorus-solubilizing halo ranged from 6.80 to 16.43 mm,with available phosphorus content ranging from 13.17 to 18.77 μg/mL.The diameter of the potassium-re-leasing zone ranged from 14.43 to 19.41 mm,and available potassium content ranged from 18.47 to 21.47 μg/mL.The results of the growth-promoting test showed that all four strains increased the ger-mination rate of lettuce,with the germination rates ranging from 78.89%to 90.00%.All strains ex-hibited significant abilities of nitrogen fixation,phosphorus solubilization,potassium release,and growth promotion.Among them,strain Y65 showed the most significant effect on biomass accumula-tion.
