萌芽前'冬枣'枝条含水率及耐寒生理指标变化分析OACHSSCD
Dynamic changes in water content and cold-resistance physiological indicators of Ziziphus jujuba Mill.cv.'Dongzao'branches prior to germination
为明确萌芽前'冬枣'枝条的含水率及耐寒生理指标对低温胁迫的生理响应,以期为阿拉尔垦区'冬枣'采集接穗提供理论参考依据.本研究于 2024 年 3 月18 日—4月15 日'冬枣'枝条萌芽前期,每日采集1 a 生枝条,测定分析其含水率、可溶性糖、可溶性蛋白、丙二醛、游离脯氨酸含量的动态变化,结合灰色关联度综合评价.结果表明:2024 年阿拉尔垦区'冬枣'在4 月10 日芽尖出现绿点,开始逐步进入萌芽期,在4月13日全部萌芽,'冬枣'枝条的含水率在 4 月 2 日—4月 10 日相较稳定,在4 月7 日含水率最低,为33.09%.在3 月24 日—4月10 日枝条可溶性糖含量逐步升高,在4 月10 日枝条可溶性糖达到最高,为 11.24%,进入萌芽期,枝条可溶性糖含量明显下降.在 3 月 18 日—4月 15 日枝条可溶性蛋白相对稳定,整体呈下降趋势,在萌芽期 4 月 10 日含量最低,为3.11 mg∕g.在4 月1 日—4月10 日枝条丙二醛含量趋于稳定,在4 月4 日枝条丙二醛含量最低,为 0.000 5 mg∕g.在 3 月 29 日—4月 10 日枝条游离脯氨酸含量呈整体下降趋势,变化幅度较小,平均游离脯氨酸含量为 1.891 mg∕g.经灰色关联度分析,萌芽前8 d(4 月2 日—4月10 日,平均温度在9.0~16.5℃之间)'冬枣'枝条含水率和可溶性糖含量整体水平较高且稳定,丙二醛含量极低,可溶性蛋白相对稳定,为'冬枣'接穗剪取的最佳时期.
To clarify the physiological responses of water content and cold resistance indicators of Ziziphus jujuba Mill.cv.'Dongzao'branches before sprouting to low-temperature stress,and to provide a reference basis for the collection of scions in the Alar Reclamation Area.During the pre-budbreak period of Ziziphus jujuba Mill.cv.'Dongzao'from March 18th to April 15th,2024,one-year-old branches were collected daily in this research.Moisture content,soluble sugars,soluble proteins,malondialdehyde(MDA),and free proline were measured and analyzed for their response to external low-temperature conditions.The cold resistance of the branches was comprehensively evaluated in combination with the grey relational degree.The results indicated that on April 10th,2024,green spots appeared on the tips of the Ziziphus jujuba Mill.cv.'Dongzao'in the Alar,and they gradually entered the germination period.All full sprouting achieved by April 13th.Branch moisture content remained relatively stable from April 2th to April 10th,and reaching a minimum of 33.09%on April 7th.Soluble sugar content gradually increased from March 24th,peaking at 11.24%in the branches on April 10th before decreasing markedly upon sprouting.Soluble protein levels exhibited overall stability but showed a declining trend;the lowest value recorded was 3.11 mg∕g on April 10th.In the branches,MDA content stabilized between April 1th and April 10th,with a minimum of 0.000 50 mg∕g observed on April 4th.From March 29th to April 10th,the content of free proline in the branches showed an overall downward trend with a relatively small change range,and the average content of free proline was 1.891 mg∕g.Through Ggrey relational analysis re-vealed that the comprehensive evaluation of cold resistance indicators was highest during the eight days before germination(Apri 2-10th;average temperature:9.0-16.5℃).The overall water content and soluble sugar levels were relatively high and stable,the MDA concentrations was extremely low,and relatively stable soluble protein levels throughout this timeframe.This study provides a scientific theoretical basis for the optimal scion collection period for'Dongzao'.
