为了建立多肉植物'柠檬玫瑰'及其嵌合体('柠檬玫瑰锦')的组培快繁体系,以'柠檬玫瑰'和'柠檬玫瑰锦'叶片为外植体,探索二者的最佳消毒方法,探讨不同浓度 NAA 与 6-BA 对不定芽诱导、增殖、生根的影响.结果表明:'柠檬玫瑰'的最适消毒组合为 75%酒精浸泡30 s,0.5%NaClO 溶液浸泡 15 min,脱菌率达到93.41%;'柠檬玫瑰锦'的最适消毒组合为 75%酒精浸泡30 s,0.5%NaClO 溶液浸泡 12 min,脱菌率为 97.07%;'柠檬玫瑰'与'柠檬玫瑰锦'诱导不定芽的最适配方为 MS+1 mg·L-1 6-BA+0.3 mg·L-1 NAA,不定芽诱导率分别为 85.43%和 75.33%,'柠檬玫瑰锦'的出锦率达到 26.00%;'柠檬玫瑰'的增殖培养基为 MS+3 mg·L-1 6-BA+0.2 mg·L-1 NAA,增殖系数为8.56;'柠檬玫瑰锦'的最适增殖培养基为 MS+3 mg·L-1 6-BA+0.1 mg·L-1 NAA,增殖系数为11.4,出锦率达到97.50%;'柠檬玫瑰'与'柠檬玫瑰锦'生根壮苗培养基的最适配方为不添加激素的 1/2MS,二者的生根率均达到 100%;'柠檬玫瑰'与'柠檬玫瑰锦'炼苗移栽时开瓶天数均为 3 d,'柠檬玫瑰'移栽成活率为 85.79%,'柠檬玫瑰锦'移栽成活率为81.16%.综上,本研究为'柠檬玫瑰'与'柠檬玫瑰锦'的快速繁殖提供了理论基础,对其产业化发展具有重要意义.

In order to establish the tissue culture and rapid propagation system of succulent plants Echeveria'Lemon Rose'and its chimera(Echeveria'Lemon Rose'prol.Variegata),the leaves of Echeveria'Lemon Rose'and Echeveria'Lemon Rose'prol.Variegata were used as explants to screen the best disinfection methods.The effects of different concentrations of NAA and 6-BA on adventitious bud induction,proliferation and rooting were discussed.The results showed that the optimal disinfection combination of Echeveria'Lemon Rose'was 75%alcohol soaking for 30 s and 0.5%NaClO soaking for 15 min,and the sterilization rate could reach 93.41%.The optimum disinfection combination of Echeveria'Lemon Rose'prol.Variegata was 75%alcohol soaking for 30 s and 0.5%NaClO solution soaking for 12 min,and the sterilization rate reached 97.07%.The optimum medium for adventitious bud induction of Echeve-ria'Lemon Rose'and Echeveria'Lemon Rose'prol.Variegata was MS+1 mg·L-1 6-BA+0.3 mg·L-1 NAA,and the adventitious bud in-duction rates were 85.43%and 75.33%,respectively.The brocade rate of Echeveria'Lemon Rose'prol.Variegata also reached of 26.00%.The optimum formula of Echeveria'Lemon Rose'proliferation medium was MS+3 mg·L-1 6-BA+0.2 mg·L-1 NAA,and the prolifer-ation coefficient was 8.56.The optimum proliferation medium formula of Echeveria'Lemon Rose'prol.Variegata was MS+3 mg·L-1 6-BA+0.1 mg·L-1 NAA,and the proliferation coefficient was 11.4,the brocade rate reached of 97.50%.The optimum formula for rooting and strong seedling medium of Echeveria'Lemon Rose'and its chimera was 1/2MS without adding hormones.The rooting rate of both reached 100%.The number of days after opening the bottle for transplanting of Echeveria'Lemon Rose'and Echeveria'Lemon Rose'prol.Variegata were 3 d.The survival rate of Echeveria'Lemon Rose'was 85.79%,and that of Echeveria'Lemon Rose'prol.Variegata was 81.16%.In summary,this study provides a theoretical basis for the rapid propagation of Echeveria'Lemon Rose'and Echeveria'Lemon Rose'prol.Variegata,which is of great significance to its industrialization development.
